Background:Neuropathic pain(NP)is a common chronic pain with high incidence,long treatment cycle and poor effect.The weakening of inhibitory signals in the nervous system is considered to be the main cause of neuropathic pain.GABA is the most widely distributed inhibitory neurotransmitter in the nervous system.By binding to corresponding GABAAreceptors on the postsynaptic membrane,GABA triggers the opening of ligand-gated ion channels,thus playing an inhibitory role.Current studies have found that GABAA receptor not only exists in the synapse,but also exists outside the synapse.Extrapsynaptic GABAA receptors has a stronger affinity with GABA,which has a more obvious influence on the excitability of neurons.It was found that the expression of the δ subunit specific to extrapsynaptic GABAA receptors in the spinal cord decreased significantly after nerve injury.At present,no studies have reported whether there is extracsynaptic GABAA receptor expressed on DRG,whether such extracsynaptic GABAA receptor changes after nerve injury,and whether these changes play a role in the occurrence and development of neuropathic pain is still unclear.Therefore,by observing the expression and changes of GABAAreceptor δsubunit in DRG of CCI rats,this study aimed to find a new approach for the pathogenesis of neuropathic.Objective: This paper aims to investigate the expression and alteration of extrasynaptic GABAA recepter in the dorsal root hanglion(DRG)of neuropathic pain.Methods: firstly,the animal model was constructed.12 male sprayed Dawley rats were divided into two equal groups.Then establish the model and use different methods to carry out surgical intervention,and finally detect the expression of GABAA recepter δsubunit protein in L4-L5 DRG.Results: the comparison between groups showed that the MWT and PWL of CCI group was significantly lower than that of Sham group.In addition,relatively speaking,GABAA receptor in CCI group δ Significantly lower.Conclusion:GABAA recepter δsubunit exists on the DRG neurons.A reduction in the expression the δ subunit of the GABAA receptor in DRG neurons were observed after CCI in rats,which may be a therapeutic target in neuropathic pain.
