Objective: To screen out and confirm Decoy ODNs which were constructed based on the TFBS of CTGF signaling downstream molecular ETS-1,NFAT and RUNX3 can downregulate the expression of liver fibrosis related gene TIMP1 and COLIα1 in rat HSC-T6 cell.Methods:(1)By literature review and bioinformatics,ETS-1,NFAT and RUNX3 Decoy ODNs were designed and constructed;(2)After ETS-1,NFAT and RUNX3 Decoy ODNs were transfected to HSC-T6 cells which were previously transfected with p GLuc-P-TIMP1,p GLuc-TRE or p GLuc-P-COLIα1 respectively,the expression of Gaussia Luciferase was checked;(3)After ETS-1 Decoy ODN was transfected to HSC-T6 cell,the expression of TIMP1 and COLIα1 was tested by Western Blot respectively.Results:(1)ETS-1,NFAT and RUNX3 Decoy ODNs could downregulate p GLuc-P-TIMP1 activity in HSC-T6 cell;(2)ETS-1 Decoy ODN can inhibit the expression of TIMP1 and COLIα1.Conclusion:(1)ETS-1 Decoy ODN can inhibit the expression of TIMP1 in HSC-T6 cell by directly downregulating the transcriptional activity of TIMP1 promotor;(2)ETS-1Decoy ODN can inhibit the expression of COLIα1,possiblely by inhibiting the expression of TIMP1.
