Correlation Of The Relationship Between Serum LncRNA SOX2OT And Oxidative Stress In Type 2 Diabetic Patients With Different Urine Albumin Excetion Rates

Objective:Analyzed the long-chain non-codingRNA SOX2OT(lncRNA SOX2OT),8-iso-prostagelandin F2α(8-iso-prostagelandin F2α,8-iso-PGF2α),and glutathione peroxide in T2DM patients with different urinary albumin excretion rates Enzyme 1(Glutathione Peroxidase 1,GPX1)level and its correlation.It aims to provide new biological markers for the early diagnosis of DKD.Methods:The subjects of this study were T2DM patients who were treated for the first time in the Endocrinology Clinic and Endocrinology Ward of our hospital.They were divided into 3 groups according to the urinary albumin to creatinine ratio(UACR):the normal albuminuria group(D1 group)40 cases,UACR<30mg/g;microalbuminuria group(D2 group)42 cases,UACR 30-300mg/g;massive albuminuria group(D3 group)40 cases,UACR>300mg/g.A random selection of 80 healthy persons who matched the age and gender of the physical examination center of our hospital during the same period were selected as the normal control group(group N).The real-time quantitative RT-PCR(q RT-PCR)method was used to detect the expression level of lncRNA SOX2OT in the serum of the above samples;the serum GPX-1 and 8-iso-PGF2αlevels were determined by enzyme-linked immunosorbent assay.Results:Serum levels of lncRNA SOX2OT in T2DM patients were lower than those in the healthy control group(P<0.01),and the level of SOX2OT expression decreased in the large urine albumin excretion rate group compared with the normal urine albumin excretion rate group and the microurinary albumin excretion rate group(P<0.05).Compared with the healthy control group,the serum 8-iso-PGF2αlevel was significantly increased(P<0.01),and it increased in the D1,D2,and D3 groups(P<0.05).Compared with the healthy control group,the serum GPX-1 level decreased significantly(P<0.01),and decreased in the D1,D2,and D3 groups(P<0.05).Pearson correlation analysis showed that serum SOX2OT levels were negatively correlated with SBP,DBP,log UACR,log Hb A1c,TG,TC,LDL-C,UA,Scr and 8-iso-PGF2αlevels(r=-0.411、-0.377、-0.744、-0.357、-0.194、-0.377、-0.465、-0.413、-0.686、-0.526,P<0.05),which were positively correlated with GPX1 level(r=0.354,P<0.05).The results of multiple linear regression analysis showed that UACR,Hb A1c,LDL-C,8-iso-PGF2α,GPX1 were independent risk factors for SOX2OT.Conclusions:Serum SOX2OT expression level may become a biomarker for the diagnosis of DKD and the severity of renal injury,and may play a role in oxidative stress-related mechanisms,affecting the occurrence and development of DKD.
