| Objective : Betatrophin is a hormone recently discovered in the human liver.So far,several homologous sequences have been found in mammalian liver,white fat and brown adipose tissue.The hormone currently plays an important role in the development of type2 diabetes,insulin resistance and lipid metabolism.Studies have confirmed that Betatrophin can improve glucose tolerance under insulin resistance.At the same time,studies have confirmed that aerobic exercise affects Betatrophin levels by changing fat content.This study mainly reveals whether aerobic exercise and adiponectin activate the LKB1-AMPK-PGC-1α pathway to affect the expression of Betatrophin and improve the insulin resistance.Methods : The experiment was carried out around the liver tissue of C57B/6 mice,divided into three groups : normal,insulin resistance and adiponectin knockout mice.Each group was given exercise and adiponectin intervention and dual intervention.Detect the expression of Adiponectin,LKB1,p-AMPK,PGC-1α,Betatrophin by three methods of Western blot,q RT-PCR and ELISA,observe the HFD changes,and then judge whether aerobic exercise has passed LKB1-AMPK-PGC-1α Pathway to improve insulin resistanceResults : 1.In the process of detecting indicators of mice in the normal group and the knockout group,the Betatrophin in the liver of the knockout group was significantly increased compared with the normal group.After exercise,adiponectin drugs and their dual interventions,the expression level of Betatrophin was significantly reduced.Aerobic exercise and adiponectin intervention can inhibit the expression of Betatrophin.2.During the comparison between the normal group and the insulin resistance group,the content of hepatic connectin in the insulin resistance group decreased significantly,while Betatrophin increased significantly.After exercise,adiponectin and its dual intervention,there was a significant decrease.3.The LKB1/PGC-1α pathway is involved in the regulation of aerobic exercise and adiponectin on Betatrophin expression Conclusion : There is a LKB1-AMPK-PGC-1α signaling pathway in mice.Aerobic exercise and adiponectin can regulate this pathway to affect the secretion of Betatrophin,thereby affecting glucose and lipid metabolism in the body to improve insulin resistance and treat diabetes. |
