Controlled Release Of Silver Ions From AgNps Using A Hydrogel Based On Konjac Glucomannan And Chitosan For Infected Wounds

As a drug carrier,hydrogel has been proved in many experiments in the past.However,in the face of practical medical problems,the clinical application of hydrogels has been proven to be effective in less research.The clinical problem of this study is the protection and restoration of infectious wound.Because the crosslinking of the hydrogel will cause the loss of antibacterial groups in many natural polysaccharides,the antibacterial properties of the hydrogels are reduced.Nanosilver particles(Ag NPs)have been proved to be good antibacterial materials,and the silver ions released by them are the main factor of antibacterial,but excessive silver ions release is harmful to cells.Based on the special porous structure of hydrogels,we crosslinked carboxymethylated chitosan(CMCS)with oxidized konjac glucomannan(OKGM)and then mixed it with silver Ag NPs with antibacterial function to form OKGM-CMCS-Ag NPs hydrogels.This hydrogel not only exhibits excellent silver ion controlled release performance but also exhibits concentration-dependent swelling rate,porosity,and strength.Cell experiment performed that L929(fibroblasts)could proliferate on the hydrogel,which showed good cytocompatibility,and the antibacterial properties of the composite hydrogel could be confirmed by co-culture with the golden grape fungus and E.coli.In addition,we found that the swelling property of the hydrogel could absorb the exudate of the wound and ensure that the wound is moist.In summary,this composite hydrogel is expected to be an excellent skin wound repair material,especially for the protection of infectious wounds.
