| Objective: To explore the expression of miR-483-5p in clear cell renal cell carcinoma and its relationship with inflammationMethods : The microarray platform of plasma from 5 patients with cc RCC preoperatively and at day 7 postoperative ly was downloaded from the Gene Expression Omnibus,in order to detect the expression of plasma miRN A;The preoperative and postoperative plasma of 12 confirmed patients were collected to verify the above results;Collected renal cancer tissue specimens and para-cancerous tissue specimens from 58 patients with renal clear cell carcinoma diagnosed pathologically.Detection of miR-483-5p expression in renal cell carcinoma and normol tissues by RT-q PCR;Construct miR-483-5p overexpression plasmid and perform gradient co-transfection in A498 cells and 786-O cells to detect the level of expression in the cells.Divide A498 cells and 786-O cells into a control group and a miR-483-5p overexpression group 24 hours after transfection for CCK-8 experiment,cell proliferation experiment,cell scratch experiment,Transwell experiment in vitro experiments.Collect the clinicopathological parameters and inflammatory indexes of 58 patients diagnosed with renal clear cell carcinoma,and explore their relationship with miR-483-5p.Results: 1.From the GEO database and the pre-and post-operative plasma of 12 confirmed patients: miR-483-5p is significantly up-regulated in plasma after nephrectomy;2.In 58 cases of kidney cancer tissues,RT-q PCR detection indicated that miR-483-5p is low in cancer tissues and is closely related to T staging(p<0.01);3.The high relative expression of miR-483-5p was negatively correlated with neutrophil-to-lymphocyte ratio(NLR)(P=0.03)and lymphocyte-to-monocyte ratio(LMR)(P=0.26).4.O verexpression of miR-483-5p can reverse the epithelial-mesenchymal transition(EMT)process and inhibit the proliferation and metastasis of renal cancer cells5.Divide A498 and 786-O cells into two groups and conduct cell function experiments after transfection.Overexpression of miR-483-5p inhibits cell proliferation,migration and invasion.Conclusion: 1.Confirm that miR-483-5p is an important plasma and tissue biomarker of cc RCC.2.miR-483-5p is negatively correlated with tumor T stage.3.Relative expression of miR-483-5p is negatively correlated with inflammation status(such as:NLR,LMR).4.miR-483-5p inhibits cancer cell proliferation and metastasis in vitro... |
