Objective This experiment aims to study the effects of propofol on the proliferation of Schwann cells and the expression of nuclear transcription factor NF-κB in vitro,so as to explore the possible mechanism of propofol’s promotion of peripheral nerve injury repair and regeneration.Methods(1)Schwann cells were cultured in vitro,control group(B)and experimental group(P)were set up in the experiment,Schwann cells in group B were cultured in complete culture medium without propofol,and Schwann cells in group P were cultured with different concentrations of propofol 20μM(P1 group),40μM(P2 group),60μM(P3group),80μM(P4 group)and Schwann cells for 6h,12 h,24h,48 h,CCK-8 method was used to observe the effect of propofol on the proliferation of Schwann cells at different concentrations and times.(2)Schwann cells were cultured with propofol with the best concentration to promote Schwann cell proliferation for 0h,12 h,24h,and 48 h,and the nucleoprotein of Schwann cells was extracted.Western blot was used to detect the expression of NF-κB P65 and p-NF-κB P65 protein in the nucleus of Schwann cells at different time periods.(3)Schwann cells were cultured in vitro,the experiment was divided into three groups: control group(no propofol and PDTC),drug treatment group(only propofol),and drug inhibitor group(propofol + PDTC).After culture of Schwann cells for 48 h in each group,the proliferation ability of Schwann cells was detected by MTT method,the nucleoprotein of Schwann cells was extracted,and the expression level of NF-κB P65 and p-NF-κB P65 protein in each group was detected by Western blot.Results(1)Compared with group B,the absorbance value(OD)of Schwann cells in groups P1,P2,P3,P4 increased,and the proliferation ability increased,P1、P3、P4(P<0.05)and P2(P<0.01);compared with group P2,The absorbance value(OD)of Schwann cells in the P1,P3,and P4 groups decreased significantly,and the proliferation ability decreased,the difference was statistically significant(P<0.01)(2)The protein content of NF-κB P65 and p-NF-κB P65 in Schwann cell nucleus gradually increased with the time of action,reaching a peak at 48 hours(P<0.01).(3)Compared with the control group,the Schwann cell proliferation of the drug group and the drug inhibitor group increased(P<0.01),and the protein expression was up-regulated,drug group(P<0.01),the drug inhibitor group(P<0.05);compared with the drug group,the Schwann cell proliferation of the drug inhibitor group was decreased,and the protein expression was down-regulated.It is statistically significant(P<0.01).Conclusion Propofol may promote Schwann cell proliferation by regulating NF-κB to promote the repair and regeneration of peripheral nerve injury.
