Objective To explore whether propofol anesthesia can affect the spatial learning and memory ability of mice through the endoplasmic reticulum stress pathway.Methods Thirty male C57BL/6 mice were randomly divided into 3 groups according to the random number table,with 10 mice in each group:propofol group(group A),propofol+4-PBA(4-PBA is an antagonist of the endoplasmic reticulum stress pathway)group(group B),blank control group(group C),by comparing the number of times the mice crossed the platform and the time of escape latency between the three groups,the effect of propofol on the spatial learning and memory ability of the mice was observed.Group A was induced with propofol18mg/kg and pumped with 73mg/(kg·h)propofol to maintain anesthesia;After pre-injection of400mg/kg 4-PBA into the tail vein of group B mice for 30 minutes,propofol 18mg/kg was induced and 73mg/(kg·h)propofol was pumped to maintain anesthesia;Group C was injected with 1ml/(kg·h)normal saline.After 2h,the three groups all stopped the administration.After24 h,the Morris water maze experiment was performed again,and the number of crossing the platform and the escape latency time were compared and recorded.Western-blot method can detect the expression of specific proteins GRP78,CHOP,and ATF4 in the endoplasmic reticulum stress pathway,and observe whether the decline in the learning and memory ability of mice is related to the activation of the endoplasmic reticulum stress pathway.Therefore,2 hours after the end of the water maze experiment,the mice were sacrificed and brain tissues were taken to prepare specimens for testing.Western-blot method was used to detect and compare the contents of GRP78,CHOP,ATF4 protein in the brain tissue of each group of mice.Results Morris water maze experimental behavioral examination results:Compared with group A,group C escape latency was significantly shorter and the number of crossing stations increased significantly(P<0.05);compared with group A,group B’s escape latency was shorter and the number of crossing stations increased(P<0.05);Compared with the two groups,there was no significant difference in behavioral examination between the two(P>0.05)Western-blot method detects the expression of GRP78,CHOP,ATF4 protein:Compared with group A,the protein expression of GRP78,CHOP and ATF4 in group C was significantly reduced(P<0.05);Compared with group A,the protein expression of GRP78,CHOP and ATF4 in group B decreased(P<0.05);There was no significant difference in the expression of GRP78,CHOP,and ATF4 between the two groups B and C(P>0.05).Conclusions 1.Propofol anesthesia can reduce the spatial learning and memory ability of mice through the endoplasmic reticulum stress pathway.2.4-PBA can improve the effect of propofol on the spatial learning and memory ability of mice by blocking the endoplasmic reticulum stress pathway.
