Objective:To study the effect of caloric restriction on ferroptosis and its pathway in liver of aged mice.Methods:Twenty-four 18-month-old male C57BL/6J mice were randomly divided into 3 groups(n=8),namely,free diet group(AL group),calorie restriction group(CR group)and SIRT1 inhibitor group(SI group)by numerical table method.The tissue sections were stained with HE and Prussian blue and the pathological changes of liver cells were observed under light microscope.ROS(reactive oxygen species)and GSH(glutathione)were determined by spectrophotometry.The expressions of ferritin,transferrin and SIRT1 were observed by Western blot.Results:The morphological and structural changes of liver cells in AL group were observed by HE staining and high power microscope.The structure of liver cells in CR group was normal.The pathological changes of liver cells in SI group were less than those in AL group.Prussian blue staining showed that there were more iron granules in the AL group,few in the CR group and few in the SI group.Compared with the AL group,the transferrin and ROS expression levels in the Cr and Si groups were significantly decreased(P<0.05);The expression of ferritin and GSH increased significantly(P<0.05);The expression of SIRT1 protein in CR group was significantly increased(P<0.05);The expression of SIRT1 protein in SI group was significantly decreased(P<0.05).Compared with Cr group,the expressions of transferrin and ROS in SI group were significantly increased(P<0.05);The expressions of ferritin,SIRT1 and GSH were significantly decreased(P<0.05).Conclusion:Calorie restriction can inhibit the death of liver iron in aged mice.
