Abstract/Summary:
Objective:To explore the expression of s Sema4D in the serum of patients with Chronic Hepatitis B and its clinical significance.Method:Select 40 newly diagnosed patients with Chronic Hepatitis B as the experimental group,Select 32 patients with Chronic Hepatitis B who have produced a virological response against the virus as the virological response group,and select 32 normal healthy individuals as the control group.Patients in the experimental group were given antiviral treatment after treatment,and collected peripheral venous blood at 0 weeks,4weeks and 12 weeks.Use low-speed centrifugation to centrifuge the peripheral blood of all groups of patients,collect their serum,and separate PBMC by density gradient centrifugation.We use automatic biochemical analyzer to detect ALT and AST,PCR method to detect HBV DNA quantification,ELISA to detect serum levels of s Sema4D,TNF-α,IFN-γ,and IL-10 in all groups,and flow cytometry to detect CD4~+T and CD8+T cell percentage in PBMC.Result:1.The expression of s Sema4D in the serum of the control group was 1475.40±187.76pg/ml,the virological response group was 1321.09±197.36 pg/ml,and the test group was1146.73±254.51 pg/ml when the treatment was 0 weeks.Compared with the control group,the expression of s Sema4D in the experimental group treated with 0 weeks and the virological response group was significantly reduced(P<0.05);Compared with the experimental group treated with 0 weeks,the s Sema4D level in the virological response group was significantly increased(P<0.05).2.The expression of s Sema4D in the experimental group after 0 weeks treatment,4weeks and 12 weeks treatment were 1146.73±254.51 pg/ml,1214.70±144.14 pg/ml,and1269.35±168.96 pg/ml,respectively.With the extension of treatment time,ALT of the experimental group gradually recovered,HBV DNA decreased,and the expression of s Sema4D gradually increased.Compared with the treatment of 0 weeks,it was significant at12 weeks of treatment(P<0.01).3.With the extension of the treatment time,the expression of TNF-αin the experimental group gradually decreased.Compared with the treatment of 0 weeks,the treatment of 4 weeks and 12 weeks of treatment were both statistically significant(P<0.05).The expression of IFN-γand IL-10 gradually increased.Compared with treatment 0W,treatment 4 weeks and treatment 12 weeks were significantly different(P<0.01).4.With the extension of the treatment time,the percentage of CD4~+T and CD8~+T cells in the test group increased significantly.Compared with the treatment of 0 weeks,the treatment for 12 weeks was statistically significant(P<0.05).5.The expression of s Sema4D in the experimental group was negatively correlated with ALT and HBV DNA(P<0.05),positively correlated with IFN-γand IL-10(P<0.05),and had no significant correlation with TNF-α(P>0.05);There was no significant correlation between the level of s Sema4D and the percentage of CD4~+T and CD8~+T cells in the experimental group before and after antiviral treatment(P>0.05)。Conclusion:1.The low expression of s Sema4D may be related to the weakened immune function in patients with chronic hepatitis B.2.sSema4D can be combined with ALT and HBV DNA as one of the important indicators of clinical prognosis in patients with chronic hepatitis B.
