Objective: To investigate the mechanism and significance of reducing lung ischemia reperfusion injury by intervening Tim-3 / Galectin-9signaling pathway in mice.Methods: Part I: C57BL/6J mice were randomly assorted into six groups to establish lung ischemia reperfusion injury models with different ischemia reperfusion time.By evaluating and comparing the degree of lung tissue injury of each group,to filter out the time of reperfusion that lung injury of mice was the most severe,and to measure the expression levels of Tim-3 and Galectin-9 molecules in the most severe lung injury group.Part II: According to whether or not mice have given recombinant Galectin-9 homologous protein,Mice were randomly assorted into four groups:(1)sham group: only to open the chest without blocking the hilum;(2)I/R2 group: to block the hilum for 1h,then to reperfusion for 2h;(3)Sham+Gal-9 group: to open the chest without blocking the hilum,and to give 60 ug recombinant Galectin-9 protein in30 minutes before surgery before surgery;(4)I/R2+Gal-9 group: to block the hilum for 1h,then to reperfusion for 2h,and to give 60 ug recombinant Galectin-9 protein in 30 minutes before surgery before surgery.By evaluating and comparing the degree of lung tissue injury of each group,and to measure and compare the expression levels of Tim-3、Galectin-9 and inflammatory factors in the lung tissues of the recombinant Galectin-9 homologous protein intervention and nonintervention.Results: Part I: The ratio of wet and dry weight,pathological changes and lung injury scores of lung tissue of mice indicate that the group of ischemia for 1h and reperfusion 2h(I/R2 group)mice had the most severe lung injury,and the ratio of wet and dry of lung tissue was7.99 ± 0.58,and the lung injury score was 0.78 ± 0.03,to compare with the other groups,showed that lung injury of alleviated(P<0.05).Moreover,the group compared with the sham group on the expression level of Tim-3 and Gal-9,the group had obviously increased on the expression level of Tim-3 and Gal-9(P<0.05).Part II: To compare the recombinant Galectin-9 homologous protein intervention with the nonintervention I/R2 group,showed that lung injury of mice were alleviated on the level of the lung wet/dry weight ratio,pathological changes,and lung injury scores of the lung tissues.Lung wet/dry weight ratios were 6.26±0.37 and 8.02±0.52 respectively(P<0.05),lung injury scores were 0.69±0.04 and 0.79±0.03 respectively(P<0.05).And the recombinant Galectin-9 homologous protein intervention group(I/R2+Gal-9 group)had obviously decreased on the expression level of Tim-3,TNF-αand IL-1β(P<0.05).Conclusion: The most severe model of lung ischemia reperfusion injury was to ischemia for 1h and to reperfusion for 2h in mice.The increased expression of Tim-3 and Gal-9 was closely related to lung ischemia reperfusion injury in the lung of mice.The Recombinant Galectin-9homologous protein could reduce lung ischemia reperfusion injury by intervening Tim-3/Galectin-9 signaling pathway in mice. |
