Wheat is an important food crop.The ear of wheat infected by Fusarium head bright(FHB)and produces a variety of mycotoxins which are difficult to degrade,In particular,Deoxynivalenol(DON),Nivalenol(NIV)and Zearalenone(ZEN)toxins,which infect wheat and its derivatives,have a negative impact on food security.Fusarium head bright resistant mechanism is complex,and more used in genetic research disease index(DI)as indicators of resistance identification,and late infection or varieties of active detoxification mechanism leads to DI with low toxins accumulation related or unrelated,direct determination of the amount of toxin accumulation in grains is more intuitive,can provide more references for gibberellic disease resistant breeding.The main research results are as follows:MFC-UPLC-DAD detection system :Simultaneous determination of DON,NIV and ZEN.Using(2.1mm×100mm,1.8μm)C18 column.The mobile phase is A:water(containing 0.1% acetic acid)and B: Acetonitrile.The gradient elution procedure is0-3min 5%B;3-9min 10%B;9-16 min 92%B;16-18 min 50%B;18-20 min 10%B.The flow rate is 0-9min 0.1ml /min;9-20 min 0.15 m L/min.The column teperature is35℃.The injection volume is 2μL.DON and NIV were detected at the diode array detector(Diode Array Detector,DAD)detection wavelength of λ=240nm.The ZEN was detected at DAD detection wavelength of λ=274nm.The results of toxin content determination of DON,NIV and ZEN in 129 wheat varieties or combinations planted in Guohe,Anhui Province in 2020 showed that: The content of DON in the same variety was generally higher than that of NIV and ZEN,consistent with that in 2019;There was a significant positive correlation between DON and NIV(r=0.619),while ZEN toxin was not highly correlated with DON and NIV(r=0.110,r=0.081).The conclusion was also consistent with that of 2019;The toxin contents of DON,NIV and ZEN in 2020 were significantly positively correlated with those of the three toxins in 2019(r=0.837,r=0.644,r=0.818),but the disease index was not correlated(r=0.075).Through the determination of toxin in wheat grains of different resistant varieties at different stages of development,the results showed: The content of DON in seeds of resistant cultivars was generally lower than that of susceptible cultivars,but NIV and ZEN did not show such a pattern.There was no obvious regularity between the contents of three toxins in wheat scab grain and the development stage of wheat..Among the 129 wheat varieties or combinations planted in Guohe in 2020,DON and NIV were significantly positively correlated with the disease index in 2020(r=0.288,r=0.462),but ZEN was not highly correlated with the disease index in 2020(r=-0.109),while the three toxins were not highly correlated with the disease index in2019.129 varieties or combinations planted in Guohe in 2020 were screened out to meet the national limit standard(DON content less than 1000μg/kg;NIV content is less than 500μg/kg;Zen content was less than 60μg/kg)in a total of 80,of which 64 varieties or combinations met the requirements for two consecutive years.The contents of three kinds of toxin in the grain were consistent with and far lower than the national limit standard for two consecutive years.Yangmai 14,Yangmai 158,Zhen Mai 9,Yang12-145,Xinong 899,Mianmai 39,Luo6073,Guinong 755,Gaoyou 2018,Bai Nong 207,Annong15-81,Y14,R146,Niavt14,Glenlen,DP34,DP36,these varieties or combinations DON content in the grain for two consecutive years is less than 350 μg/kg,NIV concentration for two consecutive years is less than 250 μg/kg,ZEN concentration for two consecutive years is less than 40 μg/kg,have a certain antitoxin accumulation ability,It can be used as the parents of antitoxin accumulation breeding.
