Sheep has high edible and use value.In recent years,the increasing demand for meat products and dairy products has directly driven the development of animal husbandry.At present,large-scale breeding has become the mainstream of sheep industry.With the increase of the density of sheep industry,the incidence of sheep disease is also increasing.Virus infection is one of the important reasons for sheep diseases.There are many kinds of ovine viruses,such as peste des petits ruminants virus(PPRV),Jaagsiekte sheep retrovirus(JSRV),Border disease virus(BDV)and so on.Once infected,it will cause large-scale disease and great economic losses to animal husbandry.In recent years,the new virus infectious disease of sheep has aroused people’s extensive attention,and the discovery and identification of new virus pathogen has become the primary task of prevention,diagnosis and treatment of viral infectious diseases.Virus metagenomics is a new virologic research method,which takes all viruses in environment and animals as research objects,and systematically reveals the virus composition when the viral genomic sequence is unknown.It plays an important role in exploring new viruses.In this study,the diversity of viruses carried by sheep was systematically investigated by virus metagenomics.After the second generation sequencing of gene library derived from 487 sheep OP samples collected in Hubei Province,34 of known viral families were found,among which viruses of 9 families infect mammals.Among these families,there are very important sheep virus pathogens,including adenovirus,parvovirus,papillomavirus,polyomavirus,picornavirus,picobirnavirus and so on.In the results of high-throughput sequencing,adenovirus had the largest number of viral reads.Through the design of specific primers,we completed the epidemiological investigation and genome sequence identification of ovine adenovirus HNU4 in sheep,and predicted that it encoded 29 genes.Ovine adenovirus HNU4 and ovine adenovirus 8 had high nucleotide sequence homology in genomic level(90.675%),and further genetic evolution analysis showed that they were the closest.They are belong to C species of ovine masteadenovirus.The results of this study revealed the genome of ovine adenovirus HNU4,and completed the whole genome sequence,genome annotation and genetic evolution analysis of HNU4,It also provides a good start for further study on the transmission mechanism of the virus and its potential harm to animal husbandry or human beings.
