Lily is a perennial bulb flower.It is from the genus Lilium of Liliaceae,and is one of the most important cut flowers in the world.China is the center of wild lily distribution,However,the most of China’s lily bulbs are imported from the Netherlands,because the cultivation of new varieties starts late and develops slowly in China.At present,the existing lily resources should be used to cultivate lily varieties with independent intellectual property rights and realize its germplasm innovation.In this study,allotriploid lily ‘Triumphator’(LLO,2n = 3x = 36)was used as experimental material.The abnormal behavior of its meiosis chromosome,development of male and female gametes,hybridization affinity,and genome composition of its hybrid offspring were analyzed.The results are as follows :1.The results of meiosis of pollen mother cells showed that the average valent configuration of ‘Triumphator’ metaphase I was 9.25/9.17 I + 11.5/11.42 II + 0.25/0.33 III+ 0.75 IV.the homoeologous chromosomes of two L-genomes formed bivalents and then disjoined and their half-bivalents moved to opposite poles at anaphase I;The chromosomes of O-genome formed univalents,among which,some attached to spindle fibers on two sides,their sister chromtids moved to the opposite poles slower than half-bivalents;some detached to spindle fibers on one side,they move to one pole at similar speed to half-bivalents;some of which deattached to spindle fibers at kinechores on either side,they became micronuclei at anaphase I.The final pollen morphology is diverse,with high shrinkage rate,but mostly ellipsoid.2.The result of anther development shows that there were six stamens in each flower,and four pollen sacs in each anther.Moreover,the degradation of tapetum and microspore development were not synchronized,and because tapetum could not normally provide nutrients for microspore development.3.Five diploid lilies ‘White fox’(LL),‘Sorbonne’(OO),‘Siberia’(OO),L.regale(TT),L.brownii var.viridulum(BB)have male fertile.The ‘Triumphator’ was used as the female parent to cross with five diploids.The fruits development have the different degrees,Only the hybridization with LL/BB obtained 14 and 40 hybrid seedlings respectively.4.Genome in situ hybridization analysis was performed on the hybrid offspring randomly,and the offspring were all aneuploid.The number of L/L + B genome chromosomes in the hybrid offspring of LLO × LL/BB was 24,and the number of O-genome chromosomes was between 2 and 8,4.L and B genome chromosomes could not be distinguished by GISH,and no recombination was observed.To sum up,allotriploid lily ‘Triumphator’ has abnormal meiosis resulting in male sterility,but can be used as female parent to cross with the appropriate diploid to produce offspring for aneuploid.This provides the clue to realize the germplasm fusion between different lily varieties of lily for breeding new lily varieties.
