Abstract/Summary:
In this study Xinjiang wild Medicago falcata L.was taken as the research object,With the analyzing the phenotypic traits and chromosome ploidy of Medicago falcata L combined with molecular markers,To explore the correlation and intrinsic relationship between the phenotype and molecular level.from the richness,morphological diversity and genetic characteristics of the germplasm resources of the Medicago falcata L.in Xinjiang,to provide a theoretical basis for further development and utilization of germplasm resources,germplasm innovation and breeding of new varieties in Xinjiang.The results as followed:There were abundant variations in plant height and stem phenotype,leaf and flower traits of wild Medicago falcata L.from different sources,and the variation range of different morphological traits was different.(1)The average variation coefficient of plant height and stem traits was 3.09%~10.13%;The average coefficient of variation of the material BEJ06(Burjin)in the main 4 traits of the leaf was the largest,which was 11.76%,and the coefficient of variation from EM03(Emin)was the smallest,which was 5.43%;The variation of flower traits is 4.67%~11.76%.Among them,the average coefficient of variation from BEJ 07 was the largest,and the coefficient of variation from HBH03 was the smallest;fruit the average coefficient of variation was between 9.17%and 18.96;(2)Correlation analysis showed that Correlation analysis showed that plant height and stem nodes,leaves,and flowers.There were significant or extremely significant correlations among the traits of fruit,indicating that the correlation analysis clearly described the close correlation between the traits of the Medicago falcata L;(3)Results of principal component analysis revealed that.The cumulative contribution rate of the first 13 principal components reached 84.83%(>80%).The experimental data further indicated that the variation of phenotypic traits among 36 materials mainly came from growth indicators and leaf width.The impact of florets,inflorescences,and seed indicators reflects most of the sources of overall data.According to 37 phenotypic traits,36 pieces of were clustered and 36 parts were divided into 3 categories at 15 Euclidean distance.The Group Ⅰ plants were low,the leaves were small,and most of the fruits were linear.Group Ⅱ plants are tall and have large leaves.The remaining materials in Group Ⅲ are of medium level.2.Thirty-six populations are divided into four ploidy levels,The results of ploidy identification indicated that 2 materials were two ploidy levels,the other remaining materials were tetraploid.(2)A total of 93 bands were amplified from the analysis of 36 wild the Medicago falcata L.materials by 9 pairs of SSR primers.The average number of primers amplified was 10.22,and the polymorphic loci accounted for 93.55%.UPGMA cluster analysis and principal component analysis of 36 the Medicago falcata L.showed that the genetic relationship of 36 Medicago falcata L.was complicated,and 36 materials were finally divided into six genetic groups.
