| In this experiment,the effect of Leucine(Leu)on the plasma amino acid and its related metabolites concentrations,antioxidant capacity and weight gain in Yili horse was explored by feeding different doses of Leu for Yili horse breeding.And the determination of Leu’s requirements provides a scientific basis.This experiment selected a single factor multilevel research method.Twenty Yili horses with similar body weight(321.68±25.7)and aged 1 years old,were divided into 4 groups with 5 horses(3♂’+2♀)in each group,Leu was not supplemented in the control group,17 g Leu was supplemented in the trial group Ⅰ,34 g Leu was supplemented in the trial group Ⅱ,51 g Leu was supplemented in the trial group Ⅲ.The trial was lasted for 21 days,on the 21st day of the experiment and blood samples were taken.The results showed that:1)Compared with the control group,the plasma Leu concentrations in the trial group Ⅲ increased by 90.53%(P<0.05),and the plasma isoleucine concentrations in the trial group Ⅰ and trial group Ⅱ decreased by 32.18%(P<0.05)and 50.53%(P<0.01),the plasma isoleucine concentrations of the trial groupⅢ decreased by 39.27%(P<0.05),and the plasma valine concentrations of the trial group Ⅱshowed a downward trend(P=0.044);the trial group I and the trial group Ⅱ Plasma glutamate concentrations decreased by 37.07%(P<0.01)and 26.52%(P<0.05).The 3-methylhistidine concentrations in the trial group Ⅲ showed a downward trend(P=0.055),and the plasma cysteine concentrations showed a downward trend(P=0.060).2)The plasma α-ketoisocaproic acid concentrations of each group gradually increased after supplemented Leu,and reached its peak at 1 h after feeding;before feeding,and after feeding 2 h,4 h,compared with the control group the plasma α-ketoisocaproic acid concentrations in trial group Ⅲ increased by 30.56%(P<0.05),43.54%(P<0.05),53.91%(P<0.01).Supplemented with Leu had no significant effect on theβ-hydroxy-β-methylbutyrate concentrations in plasma of Yili horses(P>0.05).3)Compared with the control group,the plasma malondialdehyde concentrations in the trial group Ⅲ increased by 15.69%(P<0.01),and the catalase and glutathione peroxidase activities in the trial group Ⅱdecreased by 7.62%and 4.02%(P<0.05).The plasma catalase,total-antioxidant capacity,glutathione peroxidase,and superoxide dismutase activities in the group decreased by 14.64%,17.21%,6.35%,and 11.71%(P<0.01).Supplemented with Leu had no significant effect on the plasma protein concentrations of Yili horses(P>0.05).Compared with the control group,the plasma urea nitrogen concentrations in the trial group Ⅱ increased by 58.12%(P<0.05).The impact is not significant.Under the conditions of this experiment,different doses of leucine was supplemented increased plasma Leu and α-ketoisocaproic acid levels in Yili horse and reduce plasma isoleucine levels.Leu supplemental doses greater than 34 g/horse d reduced plasma antioxidant capacity of Yili horses. |
