China is a large pig breeding country,and the outbreak of swine infectious diseases seriously affects the economic efficiency of farmers.In order to avoid the outbreak of swine infectious diseases,it is necessary for farmers to vaccinate pigs regularly,but the immunization effect of existing domestic swine vaccines is uneven,with poor immunogenicity and short duration of action,requiring multiple immunizations to achieve the desired immunization effect.In order to improve such vaccine problems,the addition of vaccine adjuvants is particularly important.In this study,Astragalus membranaceus was combined with porcine GM-CSF protein and flagellin Flic,and used as a compound adjuvant to immunize mice together with porcine circovirus vaccine,and the specific antibody levels in mouse serum were detected by Elisa assay,and the percentages of lymphocytes and macrophages in peripheral blood of mice were detected by flow cytometry to investigate the effect of cytokines and Chinese medicine compound adjuvant on vaccine enhancement The effects of cytokines and Chinese herbal adjuvants on the vaccine boosting effect were investigated.The porcine GM-CSF protein and flagellin Flic were first purified and expressed to obtain the target proteins.Then,BALB/c female mice after one week of feeding were randomly divided into 5 groups,including 1 control group,injected with phosphate solution;4 groups immunized with porcine circlet vaccine:porcine circlet vaccine alone,astragalus+porcine circlet vaccine group,GM-CSF+astragalus+porcine circlet vaccine group,GM-CSF+Flic+astragalus+porcine circlet vaccine group.Blood was collected from the tail vein on days 7,14,21,28 and 35 after immunization,and the specific antibody levels in the serum of mice were detected by Elisa method.It was found that the antibody potency of GM-CSF+Flic+Astragalus+pig ring vaccine group was the highest after immunization;GM-CSF+Flic+Astragalus+pig ring vaccine group was second only to GM-CSF+Flic+Astragalus+pig ring vaccine group,and it reached the peak on the 28th day after immunization.The antibody efficacy of each group still remained high.Blood was also collected from the eyes on days 7,14,21 and 28 after immunization,and the peripheral blood immune cell content of mice was measured by flow cytometry.The results showed that when GM-CSF,Flic and Astragalus were co-immunized with the vaccine in mice,they could better promote the differentiation of CD4~+and CD8~+T lymphocytes,and the enhancement of T cell immune effect was obvious than both the vaccine alone and the use of Astragalus+vaccine;and the immune enhancing effect of macrophages was more significant,which could rapidly promote the differentiation of CD11b~+and CD11c~+macrophages.Combined with the above experimental results,GM-CSF+Flic+Astragalus as a compound adjuvant enhanced humoral and cellular immune responses by stimulating the body to produce high levels of specific antibodies as well as rapidly promoting the differentiation of CD11b~+and CD11c~+macrophages and the proliferation of CD4~+and CD8~+T lymphocytes,using porcine circovirus vaccine as a model vaccine. |
