In order to expand the genetic basis of common potato and increase its allelic diversity,the female parent Tetraploid Potato was crossed with the male parent diploid potato by one-way sexual polyploidy,and the excellent genes of diploid potato were introduced into common potato,In this experiment,flow cytometry,chromosome counting and guard cell chloroplast counting were used to identify the ploidy of 4x-2x interspecific hybrid progenies of potato,and the main agronomic characters in the field were comprehensively analyzed and evaluated.The results show that:(1)Of 25 copies of 4x-2x interbred hybrid offspring,3 diploid,11 and11 tetraploid.The chloroplast number of guard cells was diploid in the range of 5-7,triploid in the range of 8-9 and tetraploid in the range of10-13;the perimeter of guard cells was diploid in the range of 61-70 μm,triploid in the range of 74-85 μm and tetraploid in the range of 88-104 μm.(2)The accuracy of the two methods was 100%,and that of the guard cell chloroplast counting method was 96%.(3)Six of the 25 4x-2x interspecific hybrid progenies had good characters sch as potato type,potato skin type,potato skin color,bud eye depth,potato flesh color and high yield.Three of 25 4x-2x interspecific hybrids had high starch content and high yield.(4)Among the 25 4x-2x interspecific hybrids,three were triploid zj1882.1,tetraploid zj1818.4 and zj1869.3.
