Tissue Specific Expression Of ARF,AUX/IAA And GH3 In Sweet Cherry (Prunus Avium L.) And Functional Analysis Of PavARF4

Abstract/Summary:
Auxin plays an important role in the process of plant organ development and regulation of shedding.The gene families related to auxin response include ARF,AUX/IAA and GH3.Sweet cherries(Prunus avium L.)are prone to abnormal abscising during fruit development.In this study,we searched the whole genome of sweet cherries,and obtained all ARF,AUX/IAA and GH3 members,through bioinformatics analysis,tissue-specific expression,gene cloning,subcellular localization and genetic transformation analysis functions.The main results are as follows.1.Discovery of genes related to sweet cherry auxin response.Sixteen ARF,seventeen AUX/IAA,and eight GH3 members were identified in the sweet cherry genome.Both ARF and AUX/IAA are predicted to be in the nucleus,and GH3 are predicted to be in the chloroplast.The gene structure of the same gene family is conserved and similar.It is evolutionarily similar to other species and may have extensive interactions.2.Tissue-specific expression of sweet cherry auxin response related genesTissue-specific expression results indicate that PavARF6/10/4/16,PavAUX/IAA8/10/14/21/22 and PavGH3.2/6/7 may be involved in leaf development;PavARF3/6/8/10/14/15,PavAUX/IAA8/21 and PavGH3.2/6/7 may be related to flower development;PavARF6/7/10/12,PavAUX/IAA8/16/19 and PavGH3.5 may be related to fruit development.3.Relationship between sweet cherry auxin response related genes and sheddingIn the two physiological fruit abscission,the expressions of PavARF7/8/12/13,PavAUX/IAA10/12/13/15 and PavGH3.5/6 were significantly different.These genes may be closely related to fruit abscising.4.The response pattern of genes after exogenous hormones,low temperature and drought treatmentThe expression of PavARF2/5/8/12/13/16 was up-regulated 1 h after low temperature treatment,indicating that it could respond to low temperature induction.PavARF1/5/7/10/11/16 can be induced by drought;under drought stress,the expression of PavARF1 is up-regulated.PavARF5/7/11 was down-regulated 4-6 h after treatment,and PavARF16 was up-regulated 8 h after treatment.In the PavGH3 gene family,PavGH3.2/3/6/7 can be treated with different levels of exogenous hormones.5.Gene cloning,subcellular localization and genetic transformationThe full length coding sequences of 2094 bp PavARF4,1198 bp PavAUX/IAA8,and 1080 bp PavAUX/IAA 10 were cloned,and an overexpression vector was constructed.PavARF4 is located in the nucleus and genetically transformed tobacco to obtain 6 transgenic tobacco lines;it may be related to root,seed weight,cell growth,cell wall and other life activities.
