In recent years,due to the deteriorating ecological environment and excessive and disorderly mining driven by economic interests,the number of wild apocynum has dropped sharply and is approaching endangered.At present,there is a lack of systematic research on the differences and genetic diversity of the medicinal value of wild apocynum in different regions.This article compares the chemical components of Apocynum venetum from 5 different regions,and analyzes the genetic diversity of 3 Apocynum venetum populations with extremely large geographic distances.This is to identify and effectively protect the germplasm resources of Apocynum venetum.The development and utilization of apocynum germplasm resources provide a theoretical basis.The results of the study are as follows:1.The ISSR molecular marker method was used to detect 59 polymorphic loci,the ratio of polymorphic loci was 89.4%,the Nei’s gene diversity index at the species level was 0.3267,the Shannon information index was 0.4901,and the Nei’s genes in 3populations The diversity index varies from 0.2239 to 0.2503,and the Shannon information index varies from 0.3319 to 0.3802.The total gene diversity of the 3populations is(Ht)0.3267,the genetic differentiation coefficient(Gst)is 0.2708,and the gene flow(Nm)Is 1.3463.Apocynum venetum has rich genetic diversity,and the main source of variation of apocynum germplasm resources is the variation within and between populations.2.The flavonoid content of 29 samples from 5 different wild Apocynum populations was determined.The content of isoquercitrin was between 0.28% and 0.59%,and the content of rutin was between 0% and 0.39%.The content of hyperoside is between 0.15%and 0.46%,the content of astragaloside is between 0% and 0.37%,and the content of quercetin is between 0.12% and 0.53%.The average content of the five flavonoids was in the order of isoquercitrin> hyperoside> quercetin> astragaloside> rutin.The samples from Baicheng,Jilin Province contained higher levels of isoquercitrin,astragalin,hyperoside,and quercetin,while those from Minqin County,Gansu Province contained higher levels of rutin.Principal component analysis results show that there are differences in the content of Apocynum venetum samples from different sources.17 amino acids were detected in3.Five wild Apocynum populations all detected 17 kinds of amino acids,including 8kinds of medicinal amino acids and 7 kinds of essential amino acids.The content of these two types of amino acids is the highest in the wild Apocynum populations in Minqin County,Gansu Province(9.260% and 6.114% respectively),and the lowest in the wild Apocynum populations in Hanggin Banner,Inner Mongolia(4.843% and 3.268%,respectively).The ratios of medicinal amino acids,essential amino acids and total amino acids in leaves of different apocynum populations are not much different.The ratios of essential amino acids to non-essential amino acids and the ratios of essential amino acids to total amino acids are in line with the WHO/FAO standard model.Level,the 5Apocynum venetum populations all have rich nutritional value of amino acids.
