| Salix psammophila is a unique sand shrub in Northwest China.It is an excellent tree species that prevents wind and fixes sand,adapts to barren land,and maintains the stability of the sandy ecosystem.The CCD gene family is to inhibit the formation of side branches by controlling the synthesis of solitary lactone,so this study selects a variety of Shalin,which is a diverse of branched development types to study materials,excavation of Shaow’s CCD gene family resources,and carry out the previous period of gene function.the study.It is of great value and significance for the mid-breeding of the future,and the application development of potential value.In this study,the genes of SpsCCD7 and SpsCCD8 were cloned from Salix psammophila by homologous cloning technology,And conduct bioinformatics analysis,tissue expression specific analysis,construction and transformation of target gene overexpression vector.The results are as follows:1.The analysis of the domains of the CCD gene family of the poplar family found that they all have the typical domains of the RPE65 gene family.In addition,the length of the protein encoded by the CCD gene is 353~614 amino acids,the molecular mass is between 91.68~153.51 KDa,and the theoretical isoelectric point is 4.92 Between ~5.07.The phylogenetic tree and sequence characteristics analysis of CCD gene family.Among them,CCD1,CCD2,CCD3,and CCD4 are grouped into one category,and their gene structure is the same as the phylogenetic tree.2.The CDS sequences of the SpsCCD7 and SpsCCD8 genes in Salix are 1843 bp and1674bp,encoding 614 and 557 amino acids.They are hydrophilic proteins.There are no transmembrane domains and no signal peptide regions.The phylogenetic tree grouped the amino acid sequences of SpsCCD7 and SpsCCD8 with the amino acid sequences encoded by the poplar,willow CCD7 and CCD8 genes.3.RT-qPCR results showed that the SpsCCD7 gene expressed the highest in the shoot tip and was expressed in other parts.4.The overexpression vectors 35s::SpsCCD7 and 35s::SpsCCD8 of SpsCCD7 and SpsCCD8 genes were constructed,and the poplar gene transformation work of the overexpression vectors was initially carried out.The above research results indicate that SpsCCD7 and SpsCCD8 cloned in this study belong to the CCD gene family,and their genes are related to the growth of plant lateral branches.This study further enriched the molecular genetic information of the CCD gene family,laid the foundation for the functional study of the CCD7 and CCD8 genes in Salix spp.,And provided a theoretical basis for Salix spp.Breeding work. |
