Paeonia lactiflora Pall.is a perennial perennial herb of Paeonia L.(Paeoniaceae).The dried roots of it contain a variety of active constituents displaying various biological activities such as analgesic,spasmolysis,liver protection,antihypoxic and antimalarial.Bozhou of Anhui province,panan of Zhejiang province and zhongjiang of Sichuan province are the main cultivation places for medicinal P.lactiflora.Sichuan cultivated medicinal P.lactiflora(‘Chuanshaoyao’)is of excellent quality,but it only blooms and does not bear fruit,and can only reproduce asexually.The long-term single asexual reproduction mode and only planting without breeding lead to population mixing,and severely degenerated species,and it is urgent to purify and rejuvenate.However,there are no truly confirmed(or authorized)medicinal ‘Chuanshaoyao’ varieties.Therefore,in this study,two new ‘Chuanshaoyao’ cultivars,‘Baihuachuanshaoyao’(CSYB)and ‘Honghuachuanshaoyao’(CSY-H),and the introduced ‘Hangshaoyao’(HSY)and‘Boshaoyao’(BSY)were used as materials.The differences in salt and alkali resistance and disease resistance of different cultivars medicinal P.lactiflora were compared.The aim is to further improve the study on the resistance research of the new ‘Chuanshaoyao’ cultivars(CSY-B and CSY-H),and to provide theoretical basis for the identification and promotion of new varieties of ‘Chuanshaoyao’.The results were as follows:1.Pot experiments were used to simulate salt and alkali stress.The effects of two stresses on the morphology,physiology and biochemistry of four cultivars P.lactiflora were studied,and their salt and alkali tolerance were comprehensively evaluated by the membership function method.The results showed that,with the increase of salt and alkali stress,the leaves of P.lactiflora gradually turned yellow,curled,and dried,chlorophyll synthesis and accumulation were inhibited,and cell membrane permeability increased.Under two kinds of stress,P.lactiflora can maintain osmotic regulation by accumulating a large amount of soluble sugar and proline,and improve the activity of antioxidant enzymes(SOD and POD)to alleviate oxidative damage.The salt tolerance of different cultivars medicinal P.lactiflora lines was BSY> HSY> CSY-B> CSY-H.The alkali tolerance of different cultivars medicinal P.lactiflora lines was BSY> CSY-B> HSY> CSY-H.2.The potted live plant inoculation method was used to study the resistance of medicinal P.lactiflora to gray mold.The results showed that,BSY,CSY-B and CSY-H were medium resistant cultivars,and HSY were medium sensitive cultivar.Observation found that,after infection with gray mold,the condition of HSY developed the fastest,followed by CSY-H and CSY-B,which of BSY developed the slowest.The measurement results of physiological and biochemical indicators showed that,after infection with gray mold,the PPO,PAL,SOD,POD activities and MDA content,were significantly higher than before the inoculation,and they generally increased first and then decreased,and the CAT activity finally decreased to the level before inoculation.Compared with other cultivars,the SOD,POD,PPO and PAL activitives of BSY increased more significantly after inoculation with gray mold(P <0.05).3.The potted live plant inoculation method was used to study the resistance of medicinal P.lactiflora to leaf mold.The results showed that,BSY was medium resistant cultivar,CSYB and CSY-H were medium sensitive cultivars,and HSY was high sensitive cultivar.Observation found that,after infection with leaf mold,the condition of HSY developed the fastest,followed by CSY-B and CSY-H,which of BSY developed the slowest.The measurement results of physiological and biochemical indicators showed that,after infection with leaf mold,the PAL,SOD,POD activities and MDA content increased and were significantly higher than before inoculation.The CAT activity finally decreased to the level before inoculation.Compared with other cultivars,the SOD,POD and PAL activitives of BSY increased more significantly after inoculation with leaf mold(P <0.05). |
