As a human prescription,the Chinese medicine compound"kulianmayang"injection is included in the first edition of Chinese pharmacopoeia(2015).It has the functions of relieving asthma,relieving cough and eliminating phlegm.The purpose of this study was to evaluate the effect of"kulianmayang"injection on relieving cough symptoms in terms of animal respiratory diseases.Piglets were used as target animals to evaluate its safety and its effect on relieving clinical cough symptoms.Through the above studies,it lays a foundation for the development of compound"kulianmayang"injection as a veterinary clinical cough medicine.The main research contents and results are as follows:1.Effects of"kulianyanyang"injection on the cough model induced by capsaicin in miceSpray 0.1 mmol·L-1 capsaicin solution in 0.15m L·min-1 for 3min to model,record the number of coughs of the mouse in 6min.The content of substance P in the trigeminal nerve of mice was determined by ELISA test to judge whether the modeling was successful,the number of coughs before and after modeling was counted,pathological tissue necropsy was performed after the administration,and the trachea of the drug group and model group were detected by RT-PCR The expression level of capsaicin receptor(TRPV1)in tissues was used to evaluate the antitussive effect of"Kulianmayang"injection.The results of the study showed that:(1)After the model was established,the content of substance P in the mice was detected.Compared with the blank control group,the release amount of substance P in the trigeminal ganglion of the model group had an increasing trend;(2)Each dose of"Kulianmayang"injection The number of coughs of the mice in the group after administration(9d)was significantly reduced(p<0.05),and the number of coughs in the high-dose group was the least;(3)Histopathological results showed that the blank group had normal lung tissue structure and slightly increased marginal interstitial The broad,medium-dose group had thickened lung alveolar interstitium and hemorrhage in the alveolar cavity;(4)Compared with the model group,the expression of TRPV1 m RNA in each dose group was significantly reduced(p<0.05).The above results show that"Kulianmayang"injection can reduce the number of coughs caused by capsaicin in mice,relieve the lesions caused by capsaicin in the lungs of mice,and inhibit the expression of TRPV1 m RNA,thereby alleviating the small Rat cough symptoms.2.Study on the Safety of Target Animals of"Kulianmayang"InjectionThe piglets were randomly divided into 4 groups,8 in each group.Divided into blank group,low dose group 0.148ml/kg,medium dose group 0.444ml/kg,high dose group0.74ml/kg.Intramuscular injection was given continuously for 7 days.At 0,3,and 7 days after the start of the test,the piglet’s weight,body temperature,heart rate,respiratory rate,blood routine,and blood biochemical indicators were measured;at the end of the test,the blank group and the high-dose group were dissected,and the internal organs were collected to measure the organ coefficient and make Histopathological section.The results of the study showed that:(1)"Kulianmayang"injections had no significant effect on clinical indexes such as piglet weight,body temperature,heart rate,and respiratory rate(>0.05);(2)Compared with the blank group,"Kulianmayang""Injection dose groups have no significant effect on blood routine indexes and blood biochemical indexes of piglets(>0.05);(3)Compared with the blank group,"Kulianmayang"injection is not toxic to all organs,only histology The capillaries are hyperemic to varying degrees,the parenchymal cells have no obvious lesions,and only slight granule degeneration is seen.3.Observation of Curative Effect of"Kulianmayang"Injection on Artificial Infection of Porcine contagious pleuropneumonia in Piglets.50 piglets were selected and an infectious pleural pneumonia model was established,and randomly divided into 5 groups of 10 animals each:the low-dose test group(0.074ml/kg)and the medium-dose test group(0.148ml/kg),High-dose group of tested drugs(0.296ml/kg),positive drug control group(Maxing Shigan injection 0.15ml/kg)and model group.Intramuscular injection was given for 7 consecutive days.Observe clinical symptoms,observe pathological changes,observe the improvement rate of body temperature,the improvement rate of respiratory symptoms and mortality,blood routine,blood biochemistry,organ index and other related indicators.The results of the study showed that:(1)The clinical symptoms of the model group showed decreased food intake,coughing,purulent nasal discharge,dyspnea,and increased body temperature.Each dose group of"Kulianmayang"injection and the positive drug group eased.The clinical symptom score of each dose group was significantly lower than that of the positive control group(p<0.05);(2)The results of clinical symptom improvement rate showed that the treatment effect of"Kulianmayang"injection was higher in the high-dose and middle-dose groups,and the temperature improvement rate was 100%in the high-dose group,60%in the middle-dose group,and the rate of improvement in respiratory symptoms was90%in the high-dose group and 70%in the middle-dose group;(3)The results of blood biochemical indicators showed that compared with the model group,each dose group after 7days of administration The number of medium erythrocytes and platelets increased.Platelets in the high-dose group had a significant difference(p<0.05)compared with the model group,and the blood potassium(K)content in the positive group increased significantly(p<0.05);(5).Visible through histopathological examination The pathological changes of the liver and lungs were more obvious.The lungs of the model group showed hyperplasia of alveolar epithelial cells,the capillaries of alveolar epithelial cells of the positive drug group were congested,and the alveolar wall of the treatment group showed hyperplasia Slight
