Rhizoma Coptis is a kind of Genuine Medicinal Materials,commonly used in the clinic,which has the functions of clear away heat dry dampness,defevering and detoxifying.The main source of this herb was the dried rhizome of Coptis chinensis Franch.,C.deltoidea C.Y.C.heng et Hsiao or C.teeta Wall.They are known as "Weilian","Yalian" and "Yunlian".Due to the scarcity of wild resources and the endemic species in C hina,C.chinensis,C.deltoidea and C.teeta are included in The List of Wild Plants under Key State Protection,and are classified as second-class protected plants.In recent years,Rhizoma Coptis has performed well in the treatment of diabetes,cardiovascular and cerebrovascular diseases,especially C.deltoides,which is famous for its special effect of "treating diabetes".However,there are many constraints in the production.In order to promote the healthy and sustainable development of Sichuan Coptis industry,this paper makes an investigation and analysis of the production and utilization of Sichuan Coptis.On this basis,it carries out the related work of establishing the rapid propagation system of Coptis triangularis.The results are as follows: 1.Through literature review and field research on the production and utilization of Sichuan Coptis,it is found that :(1)Since the beginning of its own literature,Sichuan has been the main production area of Coptis chinensis.Now,due to the sharp decrease of wild resources,it is mainly cultivated,with a total of 10 cities / prefectures and 28 districts / counties.There are Coptis chinensis herbs planted,with a planting area of about 150,000 Mu.The main varieties are C.chinensis and C.deltoides,accounting for 99.84% and0.16% respectively.(2)C.deltoidea(Yalian)has the best quality and the best efficacy,but the cultivation area is always small.The main reason is that the distribution area of Coptis triangularis is extremely narrow,and it has strict requirements on the habitat,and it can not be propagated by seed,only by stolon,so the propagation efficiency is low.(3)The production area of Rhizoma Coptis produced in Sichuan has rough processing,no mechanized equipment,and low production efficiency.In order to pursue high output,farmers use a large number of chemical fertilizers and growth promoters,resulting in serious diseases of Coptis chinensis,and then use a large number of pesticides,forming a vicious circle,not only polluting the environment,but also leading to a serious decline in the quality of Rhizoma Coptis.2.In view of the fact that C.deltoides can’t be propagated by seed,and its propagation efficiency is low,this paper discusses the influence of different explant parts,different sterilization time and different concentration of mercury on the e xplant treatment,the influence of different plant growth regulators and different media on its wound differentiation and proliferation,the influence of anti-browning agents on the browning degree of explants,and the influence of bacteriostatic agents on explants.The results as fellow:(1)Explant treatment stage: Among all explants,scapes perform best in the callus induction stage.The combination of disinfection time is 75% ethanol for 30 s and0.1% Hg C l2 for 10 min.The pollution rate is 55%.The basic medium for explant start-up is 6,7-V medium.The best anti-browning agent is 0.8g / L ascorbic acid Sodium,the best bacteriostatic agent is penicillin: streptomycin 5: 1.(2)Callus stage: The optimal culture condition for callus induction is: 6,7-V +1mg / L KT + 1mg / L 2.4-D,the optimal light intensity of callus is 1500 LX,the photoperiod is 10 h light culture and 14 h dark culture The best formula for callus proliferation is: 6,7-V + 0.8mg / L 6-BA + 0.5mg / L NAA.
