Expression Of Anthocyanin Biosynthesis-re Lated LncRNA In Apple Skin

Abstract/Summary:
Long noncoding RNA(lncRNA)is a kind of noncoding RNA that can play its role in the form of RNA,such as participation in biological processes such as cell cycle regulat io n,epigenetic modification,transcription regulation,etc.In this study,’Fuji’ apple with different color patterns were used as experimental materials to find the differential expression of lncRNA through high-throughput sequencing,and the biological function of lncRNA and target genes was analyzed by quantitative real-time PCR was used to verify the significa nt difference of lncRNA.On this basis,two different color patterns ’Fuji’ peel physiologica l index determination,analysis of the bag after 11 consecutive days of fruit color difference changes.At the same time,by quantitative real-time PCR,the expression difference of lncRNA and its target genes in the peels of 33 red skinned cultivars and non-red skinned cultivars,such as ’Pink Lady’,’Starkrimson’ and ’Ruixue’,was analyzed.The main results are documented as follows:(1)High-throughput sequencing of different color patterns ’Fuji’ peels resulted in 46 significantly different expressions of lncRNA,15 of which may be used as mi RN A precursors.It is predicted that 21 cis target genes and 34 trans target genes are mainly involved in metabolic process,cel ular process,flavonoid biosynthesis,photosynthes is-antenna proteins,etc.TCONS_00045933 in Blushed ’Fuji’ expression is high,in Striped’Fuji’ expression is low,its trans target base because bHLH36,and anthocyanin biosynthe s is.It is speculated that the difference between the two different color patterns ’Fuji’ peels is lncRNA-TCONS_00045933 may be related to anthocyanin biosynthesis.(2)The color change of the peel and the content of anthocyanins were measured for 11 consecutive days after ’Fuji’ apple with different color patterns were unpeached.The results showed that L* value of Blushed ’Fuji’ peel decreased by 27.10 in the 11 days after bag removal,and L* value of Striped ’Fuji’ peel decreased by 24.09.The peels of ’Fuji’ apple with two different color patterns gradually turned red,a* value of Blushed ’Fuji’ from 2.49 to 16.05,and a* value of Striped ’Fuji’ from 4.46 to 15.96.The value of b*,C*,and h° all showed a downward trend,and b* value and C* value of Blushed ’Fuji’ peel was lower than that of Striped ’Fuji’,and the change of h° value was greater than that of Striped ’Fuji’.It is proved that after ’Fuji’ apple with two different color patterns unpage,the redness of the peel and the content of anthocyanins are gradually increasing,but the content of anthocyanins of Blushed ’Fuji’ peel is greater than that of Striped ’Fuji’.(3)By quantitative real-time PCR,the expression of TCONS_00045933 in apple skin was analyzed for 11 consecutive days after ’Fuji’ apple with different color patterns were unplushed.It was found that the TCONS_00045933 expressions in Striped ’Fuji’ peels after bag removal were 1.214,2.168,1.087,1.044,1.697,1.052,2.206,1.202,1.318,1.817 times,respectively;The TCONS_00045933 expressions in apple skin of Blushed ’Fuji’ were 4.136,1.133,1.613,1.141,1.670,1.129,1.553,1.273,1.249,1.201 times,respectively.Within 11 days of unhinging,the increase of TCONS_00045933 expression in apple skin of Blushed’Fuji’ was greater than that of Striped ’Fuji’ and the relative expression of the TCONS_00045933 was strongly positively correlated with the content of anthocyanins.This further indicates that the TCONS_00045933 is related to anthocyanin biosynthesis in apple skin of ’Fuji’.(4)The expression of TCONS_00045933 and bHLH36 in 22 red skinned cultivars and11 non-red skinned cultivars was analyzed.The results showed that the higher expression of TCONS_00045933 of the 33 apple cultivars was ’Italian Early Red’,’Huashuo’,’Yanfu No.3’,and the lower expression was ’Ruixue’,’Jinling’ and ’Wanglin’.The three cultivars with higher expression of bHLH36 were consistent with the TCONS_00045933;The lower expressions are ’Jinling’,’Jinfu’ and ’Wanglin’.Most red skinned cultivars TCONS_00045933 and bHLH36 expression is higher than non-red skinned cultivars,the expression of the two genes are very significant positive correlation.This indicates that TCONS_00045933 is related to the red peel and has a co-expression relationship with bHLH36.
