With the advent of the era of total ban,it is urgent to develop a safe and healthy br eeding environment,in which the use of microbial fermented feed has become a hot sp ot.As a high-quality protein resource of mixed meal,cottonseed meal has high protein content and comprehensive nutritional components,but its effective utilization in anima l husbandry is limited because of its endogenous toxic substance-free gossypol.In this s tudy,cottonseed meal was fermented by compound microorganisms to degrade or trans form anti-nutritional factors and other harmful substances to produce many kinds of org anic acids,vitamin B and other nutrients,which can produce light fragrance in the proc ess of fermentation.improve the palatability of cottonseed meal.The test results are as follows:(1)Candidautilis,LR001 and S77 have higher detoxification rate,S77 and S007 pr oduce protease and detoxification rate is higher.Saccharomyces cerevisiae ZY.003 is ce llulase-producing bacteria,LR002 is the best lactic acid bacteria.The results of compou nd fermentation of different strains showed that the optimal fermentation strain combin ation was A(Bacillus S77)+ B(Saccharomyces cerevisiae ZY.003)+ C(Lactobacillus plantarum LR002).(2)the isolated strains S77 and LR002 have good resistance to acid,bile salt,gast ric juice and intestinal juice.Among them,S77 has better adhesion and higher sensitivit y to antibiotics.LR002 has good bacteriostatic effect on Escherichia coli and Staphyloc occus aureus.S77 is identified as Bacillus by 16 Sr DNA molecular biology and LR002 i s Lactobacillus plantarum.(3)the final gossypol detoxification rate was 91.54% by single factor experiment.Response surface regression analysis was used to obtain the best level value of each fac tor,which was 5% inoculum,fermentation time 73 h,fermentation temperature 35 ℃,r atio of material to water 0.92.Under these conditions,the fermentation experiment of c ottonseed meal compound probiotics was carried out,and the higher detoxification effe ct was obtained.The detoxification rate can reach 92.72%.(4)The fermentation products were naturally hydrolyzed for 12 hours,while maint aining a high detoxification rate,the nutritional quality of cottonseed meal was improve d to a certain extent,and the acid-soluble protein of cottonseed meal was increased by 50.94%.The contents of vitamin B1,B2 and B6 also increased by 121.68%,45.21% and42.13%,respectively,and the quality of cottonseed meal was improved.(5)In the feeding test,the addition of virus-free cottonseed meal at 25.50% was si gnificantly different from that of the control group D2(non-detoxification)in daily gai n and biochemical indexes(P>),but there was no significant difference between the control group D1(commercial grain)and the control group D1(commercial grain).The results showed that the feeding effect of virus-free cottonseed meal on mice was better.When a certain amount of soybean meal was replaced by a certain amount of soybean meal,it did not reduce the growth performance of mice,there was no significant differe nce in liver biochemical indexes,and decreased the activity o FGlutamic pyruvic transa minaseTo sum up,strains S77 and LR002 had good probiotic properties.When cottonsee d meal was fermented with Saccharomyces cerevisiae ZY.003,the detoxification rate o FGossypol was high,and the nutritional quality was significantly improved,which prov ided technical support for the research of cottonseed meal instead of soybean meal as pr otein raw material. |
