| The early pregnancy diagnosis of dairy cows is closely related to the benefit of dairy farms.The pregnant of cows are discovered earlier,the conversion period is shorter and the feeding efficiency is higher.This study speculated that there were pregnancy related factors in blood of dairy cows,and it’s in with three pathways: The interferon signaling pathways,toll-like receptor signaling pathways and complement signaling pathways.The research includes the following 3 tests:Test 1: The blood of dairy cows was collected on 18 th day of no-pregnancy;18th,25 th and 35 th day after pregnancy.The serum was separated from blood.STAT1,OAS1,MX1/2/3,and UBE1 L antibodies in the interferon signaling pathways;IRAK1,TLR2,TLR3,TLR4,TLR5,TLR9,IP10 and My D88 antibodies in the toll-like receptor signaling pathways;C1q,C1 r,C1s,C2,C3,C5,C9 antibodies in the complement signaling pathways.The above antibodies could be used to carry out specific recognition by western blotting,and detected pregnancy related factors in the blood of dairy cows.The experimental results showed that: The C1 q protein in the complement signaling pathways could specific expression in blood of day 18 pregnant dairy cows.Test 2: Referred to previous research results,the ISG15,EPF,and PAPP antibodies were used to verify the accuracy of pregnancy diagnosis in Test 1.The experimental results showed that: The diagnostic results of C1 q were consistent with those of ISG15,but were different from those of EPF and PAPP.This test indicated that C1 q antibody could be used to diagnosis dairy cows on 18 th day after pregnant.Test 3: We used the protein that can specifically bind to the pregnancy related factors in the blood of dairy cows as the monoclonal antibody,and made the colloidal gold test paper for early pregnancy detection of dairy cows.We tried to verify its accuracy,sensitivity and stability in the end.In this experiment,C1 q protein in complement signaling pathways was used as monoclonal antibody to prepare colloidal gold test paper for early pregnancy detection of dairy cows,which was used to detect blood of dairy cows with 18 th day after pregnancy.The results of tests were positive,with the detection accuracy as high as 91.7%,higher sensitivity and better stability.In summary,C1 q antibody in the complement signaling pathways could be used to diagnosis day 18 pregnant dairy cows.Thus,it could be made into colloidal gold strips for early pregnancy detection of dairy cows and applied to practical production. |
