In order to study the species,drug resistance and drug resistance gene of the pathogen of raccoon dog enteritis in Qinhuangdao,the pathogenic bacteria were isolated and identified by routine laboratory methods,and drug resistance of pathogenic bacteria in this area was detected by drug sensitivity test and drug resistance genes carried by isolated bacteria was detected by PCR technique.The research content and results are as follows:1.Collection of diseased materials and identification of bacteria: There were210 samples were collected of diseased and dead raccoon dog from different raccoon dog farms in Qinhuangdao.108 strains of E.coli and 57 strains of Salmonella were isolated.2.Detection and analysis of drug resistance: The susceptibility tests of 108 strains of E.coli and 57 strains of Salmonella were carried out with 24 kinds of antibiotics by KB method.The results showed that all 108 strains of E.coli and 57 strains of Salmonella showed drug resistance.E.coli isolates showed significant resistance to Trimethoprim,sulfamethoxazole,Sulfamethoxazole,Cefradine,sulfamonomethoxine,Lincomycin,Amoxicillin,oxytetracycline,neomycin,penicillin,doxycycline and Kanamycins,the drug resistance rate is over 66%,and the phenomenon of multiple drug resistance is serious,The most resistant species were 23,the least 4,and the most resistant species were 10-17.The resistance rates of 57 isolates to oxytetracycline,Amoxicillin,Sulfamethoxazole,Cefradine,Trimethoprim,sulfamethoxazole and sulfamonomethoxine were all higher than 90%,the resistance rate to penicillin,Lincomycin,Tilmicosin,neomycin and streptomycin was higher than 70%,Moreover,most strains showed multiple drug resistance,the number of drug-resistant strains ranged from 6 to 22,and the number of strains resistant to 16 drugs was the highest.3.Detection and analysis of drug resistance gene: The resistance genes of 108 strains of E.coli and 57 strains of Salmonella were detected by PCR.The results showed that aminoglycosides,β-lactam and fluoroquinolones resistance genes were found in E.coli and Salmonella of raccoon dog in Qinhuangdao area.The detection rates of aa DA1 and aa C4 were the highest of the aminoglycoside resistance genes carried by E.coli and Salmonella isolates,which were above 80%.The detection rates of gyr A,par C,and fluoroquinolone resistance genes were the highest,all above80%.The detection rate of SHV and CMY-2 were the highest in β-lactam,and most isolates carried multiple drug resistance genes,The results showed that the strains of E.coli and Salmonella isolated from raccoon dog in Qinhuangdao area carried the drug resistance gene seriously.
