Study On Induction Of Adventitious Bud Of Begonia×Benariensis ’BIG’ And Selection Of Clone Variation In Vitro

Begonia(Begonia×benariensis)’BIG’series,perennial herbaceous flowers belonging to the genus Begoniaceae,known as "dragon wing",commonly used for flower bed plant configuration and large-scale planting of green spaces.The"BIG" series is the first hybrid,and variety with long flowering period,large flowers and leaves and bright colors.In this study,in vitro culture was used to obtain plants with genetic traits consistent with the maternal parent.Using the range analysis method and the response surface regression analysis method to design the experiment,based on the in vitro culture of Begonia grandiflora,through the screening and optimization of different plant growth regulators,the study of adventitious bud induction was carried out and the optimal parameters were obtained.In order to further understand the series of ornamental characters,explore and utilize their favorable phenotypes to increase breeding efficiency.In this study,correlation,principal components and cluster analysis were carried out on the main ornamental characters of the three varieties of BIG series and the asexual propagation system of the first and third generation cuttings.Through the analysis of traits,screening of variant traits and analysis of variation factors in in vitro reproduction of Begonia grandiflora,the main phenotypes of variation caused by in vitro reproduction of Begonia grandiflora were determined.The research results are as follows:1.Begonia×benariensis’BIG’series of bud inductionThe best medium for petiole induction:’green leaf rose red flower type’l/2MS+NAA0.3mg/L+6-BA0.5mg/L+Inositol60mg/L,’green leaf safflower type’ 1/2 MS+NAA0.3mg/L+6BA0.5mg/LMS+NAA0.3mg/L+ Inosito160mg/L,copper leaf safflower type 1/2 MS+NAA0.3mg/L+6-BA0.5mg/L+inositol 70mg/LStem segment induced adventitious bud medium’Green Rose Safflower’ MS+6-BA0.8mg/L+2,4-D0.79mg/L+ TDZ0.12mg/L,"Green Leaf Safflower Type" MS+6-BA0.32mg/L+ 2,4-D1.13mg/L+TDZ0.37mg/L,"Copper Leaf Red Flower Type"MS+6-BA0.3mg/L+ 2,4-D0.8mg/L+TDZ0.19mg/L.2.Analysis of ornamental characters of Dahai Tong’ Big Brother The number of branches,flowers,stems,leaves,leaf area,plant height,leaf area and chlorophyll in the series were selected as the main ornamental characters,among which the plant height,leaf area and branch number could be used as the main components for the identification of ornamental characters of Begonia macrophylla;Among them,class Ⅳ traits were the worst,and the indexes were the lowest in 4 groups,and the number of branches and stem diameter were the lowest in 4 groups.3.In Vitro Screening of Begonia Variety Traits of Big Flower BegoniaVariations in the ’Bige’ series of Begonia are divided into two types:dwarf variation and leaf color variation.Leaf color variation is unstable and recoverable,which is ornamental.Dwarf variation is heritable.
