Leptin can reduce the accumulation of body fat by regulating signal transmission,reducing body feed intake,accelerating body metabolism and decomposing body fat tissue.Ghrelin increases body weight mainly by depositing fat.Overexpression of adiponectin(ADPQ)can increase lipid storage and adipogenesis.This study explored the relationship between the expression levels of leptin,ghrelin and ADPQ and fat deposition in tail,subcutaneous chest,perirenal and pericardial adipose tissues of Altay sheep at different months of age under the influence of growth rate and diet.Our aim is to investigate the correlation between leptin,ghrelin,ADPQ and fat deposition in Altay sheep.(1)Comparison among ADPQ,ghrelin and leptin expression and fat deposition in Altay sheep with different growth ratesTwelve lambs with daily gain of the last 10%and the first10%were selected from 500 3-month-old male Altay sheep and divided into slow growth group(SG)and fast growth group(FG).First,3 lambs in slow growth group and 3 lambs in fast growth group were slaughtered.When the remaining lambs were raised to 6 months old,3 lambs were selected from each group for slaughter,and the body weight changes before and after slaughter were recorded.The blood,subcutaneous chest,tail,perirenal and pericardiac adipose tissues were collected to study the blood biochemical indexes,morphological changes of fat,and the gene and protein expressions of leptin,ghrelin and ADPQ.The remaining 12 lambs were used for the next experiment.The results showed that the levels of GH and leptin in serum of 3-month and 6-month-old fast-growing group were significantly higher than those of slow-growing group(P<0.05),compared to the higher area of adipocytes than that of slow-growing group(P<0.01).The gene expression and protein expression levels of ADPQ and ghrelin in 3-month-old fast growth group were significantly lower than those in slow growth group(P<0.05);the gene expression and protein expression levels of leptin were significantly higher than those in slow growth group(P<0.05).The gene expression and protein expression levels of leptin and ADPQ in the 6-month-old fast growth group were significantly higher than those in the slow growth group(P<0.05);the gene expression and protein expression levels of ghrelin were significantly lower than those in the slow growth group(P<0.05).(2)Effect of diet on fat deposition and expression of ADPQ,ghrelin and leptin in Altay sheepBased on different diets,the 6-month-old slow growth group was divided into control group(SG-CG)and growth promoting group(SG-GP),while the 6-month-old fast growth group was divided into control group(FG-CG)and growth inhibiting group(FG-GI).The 6-month-old lambs in each group were fed for 30 days,and then were collected samples.The effects of diet on body weight,blood biochemical indexes,histocytomorphology and expression of ADPQ,ghrelin and leptin in Altay sheep were analyzed.The results showed that:the serum ADPQ and leptin contents in the growth promoting group were significantly higher than those in the control group after feeding in the slow growth group(P<0.05);the area of fat cells in the tail and around the kidney were significantly higher(P<0.05),the gene expression and protein expression levels of ADPQ and leptin in the growth promoting group were significantly higher(P<0.05);the gene expression and protein expression levels of ghrelin in the growth promoting group were significantly higher(P<0.05).The levels of serum ADPQ and leptin in the growth inhibition group were significantly lower than those in the control group(P<0.05),the area of tail and perirenal adipocytes were significantly lower(P<0.05),the levels of ghrelin gene expression and protein expression were significantly higher(P<0.05),and the levels of leptin and ADPQ gene expression and protein expression were significantly lower(P<0.05).In conclusion:in terms of growth rate,the increase of fat deposition in 3-month-old Altay sheep led to the decrease of ADPQ and ghrelin expression and the increase of leptin expression;with the increase of fat deposition in 6-month-old Altay sheep,the ADPQ and leptin expression were increased,while the ghrelin expression were decreased.In terms of diet,in the growth promoting group,the fat deposition increased,the expression of ghrelin decreased,and the expression of leptin and ADPQ increased;in the growth inhibiting group,the fat deposition decreased,the expression of ghrelin increased,and the expression of leptin and ADPQ decreased.Those results suggested that the growth rate and diet of Altay sheep are correlated with ADPQ,ghrelin,leptin and fat deposition.
