Seriphidium transiliense is a super xerophyte semi-shrub,and is one of the first species for replanting and improving degraded S.transiliense desert grassland.The storage period and storage method of seeds after harvest will directly affect the germination and emergence of seeds,and then Affect the effectiveness of its rebroadcast recovery.Therefore,this article adopts different storage time(storage 0,3,6,9,12,15,18,21,24 months),storage method [vacuum room temperature storage(-50 ~-60 KPa,15~25℃;NZ),Non-vacuum storage at room temperature(15 ~-25℃;N),vacuum zero-temperature storage(-50 ~-60 KPa,0~5℃;CZ),non-vacuum zero-temperature storage(0~5℃;C),Vacuum sub-zero temperature storage(-50~-60 KPa,-15~-20℃;FZ),non-vacuum sub-zero temperature storage(-15~-20℃;F)] and the combination of storage time and storage method.Thousand-seed weight,Water content,soluble sugar,relative conductivity,seed germination and seedling growth related indicators,explore the influence of storage time and storage methods on the quality of seeds,and combine the membership function analysis to screen the appropriate storage time and methods,with a view to S.transiliense The storage of wormwood seeds provides a reference basis.The main results are as follows:(1)The moisture content and thousand-grain weight of S.transiliense seeds under vacuum and low-temperature storage are higher than non-vacuum room temperature storage,and the moisture content of CZ seeds is significantly higher than other treatments by 26.90-142.27%,and FZ and CZ thousand-seed weights are 22.61-26.42 higher than those of NZ and N.%(P<0.05).The radicle length of N and NZ was significantly higher than that of FZ.The embryo length and dry weight of seedlings FZ and CZ were significantly higher than that of NZ and N.The germination rate and seedling emergence rate under low-temperature vacuum storage were higher than those under normal temperature storage.The content of soluble sugar in vacuum treatment is higher than that in non-vacuum treatment,FZ and CZ are significantly increased by 34.27~56.18% compared with N and C,while the relative conductivity of low temperature storage is significantly lower than that of room temperature storage,and C,CZ,F,FZ are significantly lower than N and NZ 33.15~43.56%.Comprehensive analysis showed that the seed vigor of S.transiliense was FZ,CZ,NZ,F,C,N from high to low.In short,vacuum storage is a suitable storage method for the seeds of S.transiliense.Among them,vacuum storage at subzero temperature and vacuum storage at zero temperature are more ideal.(2)With the extension of storage time,the moisture content,thousand-grain weight,dry weight of seedlings,and soluble sugar of S.transiliense seeds showed a downward trend,and the seed germination rate,germination vigor,germination index and radicle length showed a "decline-up-decline" trend.The relative conductivity is on the rise.Compared with the control,after 24 months of storage,the seed moisture content,thousand-grain weight,seedling dry weight,soluble sugar;seed germination rate,germination vigor,germination index,and radicle length decreased 0.24%~360.12%.The relative conductivity increased by 7.58%.According to the comprehensive analysis of the membership function,the storage time of S.transiliense seeds should not exceed 18 months,and the comprehensive evaluation value is greater than 0.407.In addition,the seed germination rate is 79.50%,the seedling dry weight is greater than 2.50 mg,and the soluble sugar is greater than 6.61 μg/kg.(3)The combination of storage time and storage method showed that when the seeds of S.transiliense were stored for 4 months,8 months,and 12 months,the storage methods were NZ,F,CZ,FZ,NZ,CZ,FZ,NZ,The seed quality under CZ remained relatively high,and compared with N,the increase rate of seed moisture content,thousand-grain weight,germination index,radicle length,seedling dry weight,soluble sugar,etc.increased by 18.15~153.33%.On the whole,the comprehensive evaluation values ??of FZ12,NZ12,NZ4,and FZ8 are relatively high,ranging from 0.55 to 0.76,which can be used as suitable storage conditions for the seeds of S.transiliense.In short,compared with room temperature storage,vacuum storage can alleviate the aging of S.transiliense seeds.It is recommended that the storage time of seeds should be kept within 18 months,and the appropriate storage methods should be selected according to the storage time. |
