Rhododendron is rich in color and has high ornamental value.Its breeding methods are mainly seed propagation,cutting and tissue culture,but seed propagation has the characteristics of long period and easy variation.Cultivation of Rhododendron seedlings through plant tissue culture and cutting can not only maintain the excellent seed property of Rhododendron,but also have the characteristics of high propagation coefficient and fast propagation speed.Under the guidance of plant cultivation and plantation cultivation,the effects of different rooting powder concentration and different substrate ratio on rooting of Rhododendron cuttings,and the influencing factors of explant disinfection time,primary culture,callus induction,subculture and rooting culture were discussed in this study.The results are as follows:(1)Study on Tissue Culture Technology of Rhododendron Stem Section①Experimental results show that the best sterilization condition for explants is 75%alcohol immersion for 20 seconds,The optimum sterilization conditions for R.moulmainense were 5%sodium hypochlorite disinfection 20 times minutes,aseptic water washing 5 times.②WPM+ZTO.5mg·L-1+NAA0.10 mg·L-1 is the best medium combination for the differentiation of stem segments of R.moulmainense,WPM+ZT5mg·L-1+IBA 0.2mg·L-1 followed.③WPM+TDZ0.1mg·L-1+NAA0.5mg·L-1 is the best medium for callus induction R.moulmainense stem explants,through ANOVA,Through ANOVA,auxin NAA more suitable for callus induction in stem segment of R.moulmainense than IBA.(2)Study on Tissue Culture Technology of Rhododendron Leaf①Disinfection time had a great influence on the germination rate of Rhododendron seeds.Azalea seeds were soaked in 75%alcohol for 10 s,Disinfection with 5%sodium hypochlorite at different times,R.simsi had the highest germination rate at 10 min,63.43%,The germination rate was highest at 5 min of Rhododendron molle G.Don and R.latoucheae disinfection,85.18%and 68.98%respectively.②The seed initiation of Rhododendron was significantly affected by different media,and the suitability of R.simsii、Rhododendron molle G.Don and R.latoucheae on different media from strong to weak was WPM>1/2MS>MS.The best medium for induction of germination was WPM+TDZ1.0mg L-1+NAA0.5mg·L-1,and WPM+TDZ0.5mg·L-1+NAA 0.1mgL-1.was the best medium for induction of red hair bud.③The callus was induced by the sterile leaves of Rhododendron,The induction effect of basic medium WPM on leaf callus was much higher than that of 1/2 MS and MS.medium WPM+TDZ1.Omg·L-1+NNAA1.0mg·L-1,was the best medium combination for inducing R.simsii and Rhododendron molle G.Don callus The induction rates were 57.46%and 53.33%respectively;WPM+TDZ0.5mg·L-1+NAA0.1mg·L-1 was the best medium for inducing R.latoucheae callus The induction rate was 71.11.④In callus differentiation,the effect of different basic media on callus induction and differentiation of leaves was significantly different.WPM+ZT 1.0mg·L-1+NAA 0.5mg·L-1 was the best medium for inducing differentiation R.simsii and Rhododendron molle G.Don plexus buds;WPM+ZT2.0mg·L-1+NAA 0.5mg·L-1 was the best medium for inducing differentiation culture of R.latoucheae buds.⑤The Rhododendron was obtained by analysis of variance in subculture proliferation,WPM+ZT l.Omg·L-1+IBA0.5mg·L-1was the medium with the highest effect on R.simsii subculture proliferation;WPM+ZT1.5mg·L-1+NAA1.0mg·L-1was the medium with the highest effect on Rhododendron molle G.Don and R.latoucheae proliferation.⑥Adding appropriate concentration of activated carbon and auxin to the rooting medium of Rhododendron can promote rooting.WPM+IBA2.0mg·L-1+NAA0.2mg·L-1+AC3.0g·L-1.is the best medium to induce R.simsii and R.latoucheae rooting WPM+IBA0.5mg·L-1+AC3.0g·L-1.is the best medium to induce Rhododendron molle G.Don rooting.(3)Study on cutting propagation technology①Different matrix ratio and different IBA concentration had significant difference in rooting effect.The cutting method was obtained by orthogonal test.The best IBA concentration for R.simsii cutting is 400,The best matrix is river sand:yellow soil=1:2,The rooting rate of the best combination was 63.49%,the highest number of adventitious roots was 12.4,the average root length was 4.31 cm,the longest adventitious root was 5.65 cm,the rooting index was 34.17 cm.②According to orthogonal test analysis IBA cutting effect has a significant effect.The best IB A concentration for cutting is 400,The best matrix is river sand:yellow soil:peat soil(2:2:1),The optimum rooting rate was 49.21%,The highest average number of adventitious roots is 9.3,Average root length 4.67 cm,5.38 cm,maximum root The rooting index was 21.59cm.③According to the orthogonal test and analysis,the effect of fast dipping method on rooting of Rhododendron IBA high concentration is poor,and yellow soil is more suitable for cutting of Rhododendron woolly cotton than other substrates.The optimum cutting IBA concentration of R.moulmainense is 400,the optimum substrate is yellow soil,the highest rooting rate is 52.38,the highest average number of adventitious roots is 6.32,the average root length is 2.96 cm,and the rooting index is 9.80cm. |
