| Sulfonamides residues in animal-derived foods pose a great threat to human health.In this study,two broad-spectrum recognition elements of sulfonamides were prepared,and two competitive chemiluminescence methods were established for the detection of sulfonamides residues in chicken.Firstly,a molecular-imprinted polymer was synthesized using sulfadibenz-astemplate,and a horseradish peroxidase marker was prepared.A direct competitive chemiluminescence method was established on an opaque plate to detect sulfadibenz-astemplate in chicken.The results show that the molecularly imprinted polymer can recognize 15 kinds of sulfonamides.The detection limits of the established chemiluminescence method for the 15 sulfonamides were 0.001~0.012 ng/mL.The intraday recoveries of the 15 sulfonamides in the blank chicken samples were 74.1%~93.4%,and the intraday recoveries were 79.5%~98.4%,and the coefficient of variation was 5.7%-14.3%.The simulation results showed that the specificity and sensitivity of the method for these 15 drugs were determined by the parameters of 3D conformation,structural dihedral Angle,molecular volume and surface charge distribution of core structure of each drug.Secondly,a monoclonal antibody was prepared by conjugating sulfadibenzene with bovine serum albumin(BSA)to form immunogen.A direct competitive immunochemiluminescence method was established by conjugating sulfadibenzene-HRP with sulfadibenzene-HRP on an opaque plate.The results showed that the antibody could be used for the detection of 32 kinds of sulfadibenzene-HRP drugs.The cross reaction rate ranged from 14%to 1778%,and the cross application rate of other drugs was less than 1%.The detection limits of 32 kinds of sulfonamides were 0.03-26 ng/mL.The intraday and intraday recoveries of 32 kinds of sulfonamides in blank chicken samples were 61.5%-96.4%,60.8%-97.4%,and coefficient of variation were 6.3%-14.2%.The simulation results show that the 3D conformation of each drug,the bending Angle of the 3D structure,the amount of charge in the core area and other parameters all have an effect on the binding of antibodies,and their comprehensive action determines the specificity and sensitivity of this method to them.The two chemiluminescence methods established in this study had the advantages of simple operation,high sensitivity and wide detection spectrum.They can be used for the routine detection of sulfonamides in meat tissues,which is of great significance for ensuring the safety of animal-based food and safeguarding the health of consumers. |
