Yeast glucan is derived from brewer’s yeast and has a variety of biological activities such as strengthening the body’s immunity and regulating intestinal health.However,it is insoluble in water,which greatly limits its application.After yeast glucan is sulfated modified(sGSC),its water solubility is increased and its biological activity is improved.At present,the research on its pharmacology,toxicology and clinical application is not perfect.In this study,methods such as in vivo and in vitro were used for research,and the results are as follows:1.Research on the pharmacological effects of sGSCA pharmacological experiment was carried out to explore the effect of sGSC on chicken macrophages HD 11 and Lactobacillus in vitro,to determine whether the sGSC has the effect of regulating immunity and promoting the growth of Lactobacillus.The conclusion shows that when sGSC is 0.06 mg/m L~1.00 mg/m L,the A 570 nm value is higher than that of the blank control group.As the concentration of sGSC increases,the OD value at A 570 nm also increases.Compared with the control group,it increased by 5.1%,12.8%,15.4%,16.7%and 23.1%in turn,but the difference was not significant(P>0.05),it showed that sGSC can regulate the activity of chicken macrophages in vitro,and sGSC may have the effect of regulating the immune function of chickens.Through the effect of sGSC on Lactobacillus,the results showed that compared with the control group(0 mg/m L),when the concentration of sGSC was 0.56 mg/m L,there was a growth-promoting effect but not significant(P>0.05).When the concentration of sGSC is in the range of 1.13~9.00 mg/m L,the pro-growth effects are all significant(P<0.05),but each group added with sGSC has different degrees of pro-growth effects on Lactobacillus.In the range of 0.56~9.00 mg/m L,with the increase of sGSC concentration,the pro-growth effect first increased and then decreased,and the pro-growth effect on Lactobacillus had the best effect at 2.25 mg/m L.2.Study on the toxicity of sulfated yeast glucanForty Kunming mice weighing 17-19 g(half male and half female,reared separately).The sGSC group(5 mg/g.BW)and the control group were given 20 mice in each group,and the mice were adapted to the environment without any stress response,and then the experiment was carried out.During the glucose observation period,each group was clinically recorded every day for 14 days.Record the weight of each mouse on the 1 d,7 d,and 14 d.After the sugar was given,the behavioral performance,mental state and death of the mice were monitored every day.After the experiment,the mice’s body weight changes,organ index changes,and organ pathological changes were analyzed.The results showed that when the concentration of sGSC was 5 mg/g.BW,it had no effect on the body weight and organ index of the mice(P>0.05),and there were no pathological changes in the organs and tissues of the mice.3.The effect of sGSC on the intestinal flora of antibiotic-associated diarrhea(AAD)miceForty mice were randomly divided into four groups:A,B,C,and D.Group A was the normal control group,and group B was the sGSC control group.The mice in groups A and B were given RO water(Reverse osmosis water,C and D groups were intragastrically administered lincomycin hydrochloride at a dose of 3 mg/g.BW.After 3 consecutive days,mice in group B and the other group of mice receiving lincomycin hydrochloride(group D)were given sGSC at a dose of 100 mg/kg.BW for14 days.The body weight,organ index,SCFAs(short-chain fatty acids)and intestinal flora changes of mice were analyzed.The results showed that sGSC is beneficial to promote the weight recovery of AAD mice and reduce the pathological changes of the ileum and colon.It can significantly increase the abundance of Lactobacillus and Coprococcus(P<0.05),and reduce the abundance of Mucispirillum and Bilophila,but the difference is not significant(P>0.05);it can promote the production of butyric acid and propionic acid in short-chain fatty acids.The results showed that sGSC has a certain therapeutic effect on the diarrhea symptoms of AAD mice,and has the function of regulating the flora in the intestinal tract of AAD mice.4.Control effect of sulfated yeast dextran on chicken coccidiosis200 chickens were randomly divided into group A low concentration group(4 mg/kg.BW),group B medium concentration group(16 mg/kg.BW),group C high concentration group(64 mg/kg.BW),group D Infected group,E group healthy group.3 days before infection with coccidiosis,the chickens of groups A,B,and C were fed with the above-mentioned concentrations of sGSC according to their body weight every day,and the chickens of groups D and E were fed with pure water(RO water)until the end of the experiment.During the experiment,the behavioral performance and bloody stools of the chickens were observed.On the 5 th and 6 th day,the feces were kept,and the total amount of oocysts per gram of feces was counted.All the chickens were killed on the 7th day after infection,and the heart,liver,spleen,Thymus and bursa index are used to evaluate the effect of sGSC on coccidiosis infection.The results showed that sGSC can improve the impact of Eimeria tenella infection on chickens and has an anti-coccidial effect.
