Under natural environment,the stress of high salt,drought,extreme temperature and diseases and insect pests will affect the growth and development of plants,and the yield and quality of the crops will reduce.Cauliflower belongs to the cruciferous Brassica genus,the main edible organ is curd,so cauliflower has very low adaptability to various adversity,and the yield and quality are seriously affected in the process of production.ERF(Ethylene responsive factor)is a kind of plant specific transcription factor.It belongs to the AP2/ERF transcription factor family.It plays an important regulatory role in plant growth and development and stress response.Reffer to the research achievement related to resistance of Arabidopsis and other plants,two ERF genes were cloned from cauliflower,the expression and function of two genes of ERF023 and ERF114 were in preliminarily explored by using molecular biology erperimental method.The results of the study laid an important foundation for revealing the function and mechanism of two ERF genes in the resistance process of cauliflower at the molecular level.The main results were as follows:1.The full length cDNA of ERF023 and ERF114 was successfully isolated and cloned from cauliflower.2.The expression characteristics of ERF023 and ERF114 in cauliflower seedlings under drought and salt stress treatment were analyzed by qRT-PCR technology,the results show the expression of cauliflower ERF023 gene increased under drought stress and salt stress,ERF114 gene expression increased under drought stress,however the expression decreased change under salt stress.3.The plant expression vector of cauliflower ERF023 and ERF114 was successfully constructed.4.ERF023 and ERF114 were transformed into Arabidopsis by Agrobacterium tumefaciens,and the transgenic Arabidopsis was screened.The function of overexpression of ERF023 and ERF114 in Arabidopsis thaliana was analyzed.The results showed that ERF023 could promote plant growth,make the transgenic plants higher,the main stem stout and the leaves bigger,and the transgenic plants showed stronger resistance to salt and drought stress,The overexpression of ERF114 in Arabidopsis led to the dwarf plant,the smaller leaf,,and the transgenic plants showed stronger drought resistance,but did not respond to the salt stress.5.The transcriptional sequence analysis of ERF114 transgenic and wild Arabidopsis plants was carried out by transcriptional sequencing.The results showed that 2,3836 genes were detected,including 1,801 differentially expressed genes,of which 1,222 were upregulation and 579 were down-regulation.The analysis of GO and KEGG enrichment of different genes showed that overexpression of ERF114 had an important regulatory effect on the growth and drought resistance of transgenic Arabidopsis plants.6.,ERF023 and ERF114 gene overexpression cauliflower plants were obtained by Agrobacterium mediated infection with the hypocotyl method of cauliflower... |
