| Solenaia oleivira is a rare and rare species of freshwater oysters in China.It has a typical life history of mites.After the mature larvae are discharged,they need to be parasitized on suitable host fish in order to metamorphose into juveniles,thus completing the breeding process.Due to the special reproduction and habitat characteristics of the olive carp,the information on the specific host fish is still unclear,and no breakthrough has been made in artificial breeding,which greatly restricts the protection,development and utilization of this rare scorpion resource.In this paper,the characteristics of reproductive biology were systematically studied by collecting the Huaihe Solenaia oleivira month by month.The in vitro culture technique of the larvae of the larvae was carried out,and the serum types and densities of the culture were screened.Seasonal changes in composition were analyzed.The key technology research of artificial breeding of Solenaia oleivira provides new green ecological breeding species for the development of modern fishery in China,and provides strong technical support for the artificial breeding protection and development of Chinese Solenaia oleivira.The main results of this study are as follows:(1)The reproductive biology study of the Solenaia oleivira showed that the age of the Solenaia oleivira was mainly from 2 to 6 years old,and the age of the first sexual maturity was 31.25 g,the shell length was 10.54 cm,and the maximum mature adult was 5 Age,weight 152.63g,shell length 18.45cm.The breeding season of the Huaihe Solenaia oleivira is from February to June each year,and the breeding peak is from the end of March to May.The germ cell development of this carp is also carried out in batches.Females can ovulate 3 to 5 times during the breeding season,and the amount of larvae is 13.49 million to 40.21 million.The amount of female larvae is positively correlated with the weight of the individual..According to the results of tissue section on the development of germ cells and changes in follicles,it is considered that the proliferative phase of gonads is from September to November,and the gonadal growth phase is from December to January of the(2)second year,from February to May.For the stage of gonad maturation,the gonad discharge period from March to June,and the gonad rest period from July to September.The olive scorpion cells and the mature sperm are stacked together and connected by flagella;the sperm is elliptical short rod-shaped,with a slender flagella at the tail,the overall length is 46~50μm,and the head is bullet-shaped,the size It is about 0.72×1.43μm.The embryonic development process of the Solenaia oleivira is divided into fertilized egg,cleavage stage,blastocyst stage,primitive intestinal stage,intramembranous larvae and larvae.(3)The results of in vitro culture showed that the highest rate of morphological changes in the larvae of the larvae was 86.61±4.42%;the lowest morphological group of the larvae was rabbits.One-way analysis of variance showed that the metamorphosis rate of the larvae of the control group was significantly lower than that of the larvae of the larvae of the serogroup(P<0.05);the rate of larvae of the larvae of the carp,the carp,the grass carp,the black carp and the cattle group.There was no significant difference(P>0.05).The morphological rate of larvae in the carp,grass carp and rabbit group was significantly different(P<0.05).The carp was higher than the metamorphosis rate of grass carp,and the grass carp was higher than the rabbit metamorphosis rate.Using squid serum as an important nutrient source,the allergic rates obtained under the conditions of in vitro medium density of 800,1200,1600,and 2000 were 36.70 ± 11.67%,58.85 ± 2.73%,68.31± 15.70%,and 78.48± 9.31%.(3)The seasonal variation analysis of the nutrient composition of the Solenaia oleivira showed that the crude ash of the Solenaia oleivira in March was significantly lower than that of June,September and December(P<0.05),and there was no significant difference in the coarse ash in June,September and December(P>0.05);The crude protein of olive mites was significantly higher in March,September and December(P<0.05);there was no significant difference in crude fat in March,June,September and December(P>0.05).The 17 kinds of amino acids contained in the Solenaia oleivira accounted for 54.31%of the total amino acids,of which the essential amino acid content was 21.93%,and the umami amino acid was 19.35%.There was no significant difference in amino acid content with seasonal changes. |
