| Selenium(Selenium,Se)is an essential trace element for the human body.Because it is extremely rare in nature,it is also called a rare element.Selenium exists in many forms.Common inorganic selenium is selenate and selenite.These two forms of inorganic selenium are absorbed by plants and transformed into organic selenium compounds through the plant’s own metabolism,mainly in a variety of organic forms.The form of selenium is stored in plants.Modern medicine shows that the lack of selenium can cause dysfunction of many important organs of the human body,and then cause disease.In many cases,it is necessary to artificially supplement selenium.There are two ways to supplement selenium in the human body: direct supplementation of inorganic selenium and consumption of agricultural products containing organic selenium.However,supplementing selenium with inorganic selenium has a narrow safety range and is likely to cause harm to the human body.In recent years,selenium supplementation has been supplemented by organic selenium.Has been widely welcomed.This study took mung bean sprouts as the research object.In order to obtain high-quality organic selenium,various selenium application methods were used to cultivate mung bean sprouts.The selenium-enriched mung bean sprout cultivation method and organic selenium extraction conditions were optimized by investigating the selenium absorption effect and the organic selenium content during the growth of mung bean sprouts.Obtain the organic selenium solution with the highest organic selenium content,and prepare the corresponding amount of inorganic selenium solution to prepare the selenium-containing nutrient solution(Se-nutrient solution)for cultivating wheat seedlings and the selenium-containing medium(Se-LB medium)for culturing bacteria.The bioavailability of organic selenium and inorganic selenium was evaluated by analyzing the growth of wheat seedlings and the changes of bacterial colonies,and the results were as follows:1.This experiment uses three methods of selenium application to cultivate mung bean sprouts.The first method is to use selenium-enriched medium for continuous cultivation for 72 hours;the second method is to use selenium-enriched medium for continuous cultivation for 24 hours and then change to ultrapure water to continue.Cultivate for 48 hours;the third method is to replace the equal-concentration selenium-enriched culture medium every day for three days.Use microwave digestion hydride atomic fluorescence method to investigate the content of total selenium,water-soluble selenium,acid-soluble selenium and alkali-soluble selenium in selenium-enriched mung bean sprouts.The first,second and third selenium application methods in mung bean sprouts The highest total selenium content was12638 μg/kg,9721 μg/kg and 22363 μg/kg,and the leaching effect was the best under alkaline conditions,and its content reached 4915 μg/kg,4718 μg/kg and 6777 μg/kg,respectively.kg,indicating that the third selenium application method is better than the first two.The experiment determined that the organic selenium content of the third selenium application method and the extract obtained under alkaline conditions was the highest.2.Use the above extract to compare with the inorganic selenium solution of the same concentration,and configure the Se-nutrient solution to cultivate wheat seedlings at room temperature with sufficient sunlight.The results showed that the leaf length,root length and chlorophyll growth indexes of wheat seedlings applied with selenium were better than the blank group,and the organic selenium group was significantly better than the inorganic selenium group.When the leaf length of the inorganic selenium group was inhibited,the organic selenium group was significantly better than the inorganic selenium group.The leaf length of wheat seedlings in the selenium group continued to increase,indicating that organic selenium has better biological efficacy and higher biological safety than inorganic selenium.3.Use the above extract to compare with the inorganic selenium solution of the same concentration,prepare Se-medium under aseptic conditions,and LB medium as a blank control experiment.Cultivate E.coli and E.coli at 28℃,37℃ and 39℃.Staphylococcus aureus.The highest concentration of organic selenium and inorganic selenium at each temperature within 4 hours can inhibit the growth of Escherichia coli and Staphylococcus aureus to a certain extent,and the inhibitory strength of the organic selenium group is almost higher than that of the inorganic selenium group.The inhibitory ability gradually weakened with the increase of time,and the growth was almost the same as that of the LB medium group after 4 hours.After 24 hours,the growth of the organic selenium group showed a trend of first promotion and then inhibition,and the growth of the inorganic selenium group showed a trend of first inhibition,then promotion and then inhibition,indicating that the bioavailability and safety of organic selenium was higher than that of inorganic selenium. |
