In this paper,12 kinds of wine grapes planted in the grape production area of Eastern Hebei Province were selected as the research objects.By investigating the damage indexes of the Apolygus lucorum to the leaves of different varieties of wine grapes and the change of the number of overwintering eggs,the excellent wine grape varieties with resistance to the Apolygus lucorum were selected.GC-MS was used to detect the volatiles released from the leaves of different varieties of grape seedlings and Cabernet Sauvignon(middle resistant variety)seedlings under different treatments.The differences of species and content were identified,and the effective components which could avoid or attract the Apolygus lucorum were predicted.In addition,the stability of internal reference genes in different treatments of Cabernet Sauvignon samples was analyzed,and the most stable internal reference genes were screened out.It provides a basis for breeding wine grape varieties resistant to Apolygus lucorum and producing new green insecticidal agents mainly composed of plant secondary metabolites.The specific research results are as follows:1.By investigating the damage indexes of Apolygus lucorum to the leaves of different wine grape varieties and the egg setting of overwintering eggs on different wine grape varieties,it shows that there are significant differences in the resistance level of different wine grape varieties to Apolygus lucorum,among which Marselan was the high resistant variety,chilongzhu,Vidal Blanc,Viognier,Italian Riesling,Petit Manseng and Cabernet Sauvignon were the middle resistant varieties,which had higher resistance to the Apolygus lucorum,and the Apolygus lucorum overwintering there was no significant correlation between the overwintering eggs of the Apolygus lucorum and the damage indexes of different wine grape varieties.2.Through GC-MS analysis of the volatiles released from the leaves of different wine grape varieties at the seedling stage,53 components were identified,included esters,hydrocarbons,aromatics,acids,ketones,alcohols,terpenes and other compounds.The results showed that phenol,cis-2-Penten-1-ol and longifolene and other compounds were unique to the highly resistant plants,while Dimethyl phthalate,n-tridecane,etidronic acid and other compounds were unique to the highly sensitive plants,which might affect the feeding behavior of the Apolygus lucorum.3.Through GC-MS analysis of the volatiles released from leaves of Cabernet Sauvignon seedlings under different treatments,31 components were identified,including fatty acid derivatives and benzene derivatives.The results showed that there were different kinds and contents of volatile substances released from Cabernet Sauvignon leaves under different treatments,among which n-tridecane,dibutyl phthalate and diisobutyl phthalate,hexyl butyrate,4’ethylacetophenone and 2,2-dimethylheptane and other compounds might be the substances that affect the resistance of Cabernet Sauvignon after being injured.4.According to the comprehensive analysis results of geNorm,NormFinder and BestKeeper,showed that GAPDH was the most stable internal reference gene and 18 S rRNA was the most unstable internal reference gene under the three treatments of mechanical damage and Apolygus lucorum damage and control. |
