R.odorata var.gigantea is a wild species of Rosa L.with excellent characteristics such as tea-scent,low-temperature flowering,which is a good material for studying the specific biological characteristics of Rosa plants.At present,there are few studies on genes related to tea-scent and flowering of R.odorata var.gigantea,which restricts the breeding and utilization of related traits.For this reason,we conducted transcriptome sequencing on eight tissues of R.odorata var.gigantea including flower bud,leaf,flower bud tip,flower,stem,fruit,stem tip and root.We through gene annotation,DEGs analysis,STEM analysis,ASE analysis and trait-related gene analysis,dig out some candidate genes that may be involved in the formation of tea-scent and flowering.Below are crucial conclusion findings:1.Constructing the transcriptome database of different tissues of R.odorata var.gigantea by using the high-throughput sequencing Novaseq platform.A total of398,698,533 clean reads were obtained,and all the alignment rate was more than 90%.DEGs analysis found there are the least differentially expressed genes between stem tip,flower bud tip and flower bud.There are 2101 differentiaily expressed genes between flower bud and flower bud tip,1269 and 832 for up-regulated/down-regulated genes;1724 differentially expressed genes between flower bud and stem tip,625 and 1099 for up-regulated/down-regulated genes;1415 differentially expressed genes between flower bud tip and stem tip,264 and 1151 for up-regulated/down-regulated genes.2.Meristem-specific gene analysis showed that three transcription factor families have been identified only in the flower buds,including MIKC-MADS(SOC1/AGL20,AGL24),BES1(BMY7)and NF-YC(NF-YC10),and two transcription factor families have been identified only in the shoot tip and flower bud tip,including AP2(TOE1)and ARR-B(ARR12).3.STEM analysis and GO analysis found that genes related to tea-scent are mainly related to metabolic processes of many compounds such as sugar and lipid;genes related to floral evocation are mainly related to various organic metabolism,cellsignaling,cell structure synthesis,various enzyme activities and transcription factor activities;genes related to flowers organ development are mainly related to cell division,cell structure synthesis,multiple organic compounds metabolism,multiple enzyme activities and transcription factor activities.4.ASE analysis found that allele specific expression is widespread in tissues of R.odorata var.gigantea.5.Bioinformatics analysis indicate that FaERF6,FaERF25,RrOOMT,RrAADC,FaLOX,RrLIS,RhNUDX1_JQ820249 and RrPAR may be involved in the formation of tea-scent of R.odorata var.gigantea;and RoAP1/FUL-like,RoEMF2,RoLHP1,RoGA20 OX,RoSPINDLY,RcAGL15,RoLFY,RoFD and RoSUF4 may be involved in the flowering regulation of R.odorata var.gigantea. |
