Circular RNA(circ RNA)is a special kind of RNA molecule which present as a circular structure.It is widely exists in various organisms,and can regulate gene expression.Cir RNAs have an important role in regulating growth and proliferation of muscle cell.In order to explore the effect of circ RNAs on the muscle growth of domestic chickens,we identified a high expression level circ RNA(named circ ZBTB10)by previous RNA sequencing of chicken primary myoblast cells.We used q RT-PCR to identify the expression pattern of circ ZBTB10 on differential kind of tissues.We found that circ ZBTB10 was highly expressed in heart,liver,kidney,breast and leg muscle of Xinghua chicken.Moreover,we investigated the effect of circ ZBTB10 on the proliferation of DF1,ICP,LMH and primary myoblast cells by overexpression and knockdown of circ ZBTB10.We investigated the function of circ ZBTB10 on cell cycle progression by q RT-PCR,flow cytometry and Ed U.We found that circ ZBTB10 promoted the cell cycle progression of chicken cells by regulating the expression of Cyclin B2,Cyclin D1,Cyclin D2,PCNA,CDKN1 B,p21 and IGF1 R.Circ ZBTB10 also had a different expression level at different stage of myoblast differentiation.Circ ZBTB10 could promote the expression of Myo D,MYOG,My HC and Myomaker genes which are related to myoblast differentiation.We utilized a prediction algorithm to identify the open reading frame of circ ZBTB10.Circ ZBTB10 may encode a 190 aa protein.We constructed an overexpression vector of circ ZBTB10 with a flag tag,and some control vectors.The western blot results identified that circ ZBTB10 could not encode the predicted polypeptide ZBTB10-190 aa.In summary,we found circ ZBTB10 could promote cell cycle progression on chicken cells,and explored the protein-encoding ability of circ ZBTB10.This study provided a new theoretical basis and direction for further study of the effect of circ RNA on growth and development of domestic chickens.
