| The establishment of molecular marker-assisted breeding system for important traits in melon has very important application value for enhancing the efficiency of breeding and germplasm innovation.More recently,melon breeding of China is in the traditional stage,which is laborious and long-term.The aims in this study is to establish a set of efficient,accurate and economical molecular marker-assisted breeding systems for important traits in melon,which includes the development and application of molecular markers for monoecious,the application of molecular markers in foreground and background of monoecious and the optimization of molecular marker-assisted selection procedure.The major research results are as follows:1.In Del-1,In Del-5 and SNP-KASP markers linked to the monoecious were developed.Genotypys of 25 accessions with different sexual phenotypes were detected using three molecular markers.The results showed that the phenotypes were consistent with genotypes.In Del markers were applied to 8 crossing of F2 populations,which could identify monoecious from different sources.The coincidence rate was above 94%,and the accuracy was high.2.The selection of monoecious foreground and background in the F2,F3and BC1F3populations from the crossing between Pa631and Pa706 was used to select new germplasm materials.The trait of monoecious in Pa706 was transferred to the Pa631.Pb549-16 line was the most similarity to the male paternt,which was back crossed with Pa631.The male paternal genome of the backcross populations were 73-91%,and new germplasm materials were selected efficiently.3.Molecular marker-assisted breeding system for important traits in melon was established by optimizing the DNA extraction and detection of multiplex PCR in melon seeds and cotyledons.This system was applied to the polymerization of traits breeding(monoecious and the orange flesh)as well as the detection of seed purity.A efficient molecular detection platform was builded.The In Del and SNP-KASP markers linked to the monoecious were developed.The molecular marker-assisted breeding technology was applied to the germplasm innovation of melon.The procedure of molecular marker assisted selection was optimized and a set of efficient,accurate and economical molecular marker-assisted breeding systems for important traits in melon was established,which was applied to the polymerization of traits breeding(monoecious and the orange flesh)as well as the detection of seed purity. |
