| Gene editing technology has been widely used in crop genetic improvement and greatly improved breeding efficiency.In order to create a new temperature-sensitive genic male sterile line through CRISPR/Cas9 gene editing technology,this study uses 6 excellent japonica rice:ZG75S,CYGS,YG0618S,ZG07S,T0361S,7679S and 4 excellent indica rice:2537S,6150S,6379S,1109S as the editing object,and the temperature-sensitive genic male sterile lines of japonica and indica were created by knocking out the TMS5 gene.By comparing the temperature of infertility of temperature-sensitive genic male sterile lines,it was found that the temperature of infertility of japonica-type temperature-sensitive male sterile lines ZG75S,CYGS,YG0618S,ZG07S,T0361S,and 7679S are all between 28℃ and 32℃,and The temperature of infertility of indica-type temperature-sensitive genic male sterile lines 2537S,6150S and 6379S is between 24℃~28℃.The temperature of infertility of 1109S is lower than 23.5℃,which has great application value.The results showed that the temperature of sterility of tms5 mutants obtained from different genetic background materials was different.It is possible to obtain temperature-sensitive nuclear male sterile lines with lower temperature of sterility by selecting TMS5 knockout from materials with different genetic background.In order to study the combining ability of low infertility starting temperature 1109S,it was tested and matched with 7 high quality male parents 8025,8028,8034,8038,8048,Huazhan and R534.Through field phenotypic identification and agronomic traits investigation,a high-yielding hybrid rice combination 1109S/8048 with strong heterosis was selected.The 1109S/8048 combination increased yield by 13.1%compared with the regional test Fengliangyou 4 and compared with the male parent the 1109S/8048 combination increased output by 25.6%.The rice quality analysis showed that there was no significant difference between the rice quality of the 1109S/8048 combination and the rice quality of the 8048.The creation of the temperature-sensitive genic male sterile line 1109S and the high-yielding hybrid rice combination 1109S/8048 provided a new approach for high-yielding breeding of hybrid rice. |
