Abstract/Summary:
Cytochrome P450 is a special family of hemoglobin(hemoglobin)characterized by enzyme activity and its role in drug metabolism.There are more than 20 enzymes in cytochrome P450.The most important enzymes are CYP1A2,CYP2E1 and CYP3A4.Most of them are also involved in the metabolism of endogenous substances,such as steroids.The activity of the enzyme can be stimulated(enzymatically induced)or inhibited(enzyme inhibited).Many drugs can increase or decrease the activity of various CYP isoenzymes by inducing biosynthesis of isoenzymes(enzyme induction)or by directly inhibiting CYP activity(enzyme inhibition).This experiment selects the potential risks of florfenicol and doxycycline veterinary antibacterial agents commonly used in the breeding industry during clinical use,and explores the effects of the combination of the two drugs on the activity of CYP450 enzymes in order to meet the specifications in animal production.use.This study explores the effects of florfenicol and doxycycline on the liver enzyme activity of chickens,and provides a basis for rational use of veterinary medicine.Bailanhang chickens were divided into two groups:florfenicol(F,200 mg/L),doxycycline(D,300 mg(L),combination(F+D),and negative control(Group C).In this experiment,blood was collected and the liver was separated after multiple administrations,and stored at-80℃ for use.The expression level of CYP mRNA in chicken liver was determined by qRT-PCR.The protein content was determined by BCA method and the physiological and biochemical indexes of chicken were determined.The experimental results showed that the florolicol in each experimental group had potential liver tissue damage compared with the normal group.The CYP450 protein concentration showed different changes,and the florfenicol group significantly increased the protein concentration.There was no significant effect on the co-administration group or the like.The florfenicol group significantly inhibited the expression of CRA factor,and the florfenicol and doxycycline groups showed significant expression of CYP3 and CYP2 genes.The results of this study indicate that the combination of florfenicol and doxycycline can reduce the potential inflammatory response in liver tissue.The combination of the two drugs inhibits the expression of CYP2D and CYP3A by inhibiting the activation pathway of CAR to avoid liver damage.The experimental results suggest the rationality of the combination of the two drugs.
