Studies have shown that there is a large difference between Polyporus umbellatus in Changbai Mountain and Polyporus umbellatus in Shaanxi province.The culture characteristics of mycelia of Changbaishan Polyporus umbellatus and Shaanxi Polyporus umbellatus are also very different under pure culture conditions.In order to understand the gene expression characteristics and provenance differences between the hyphae of the Changbai shan Polyporus umbellatus and Shaanxi Polyporus umbellatus,Illumina sequencing technology was used to sequence the 6 samples of Changbaishan and Shaanxi Polyporus umbellatus mycelium,and the differentially expressed genes of the two myceliums were compared and analyzed.A total of 133662 Transcripts and 20999 Unigenes were obtained by data quality control of the transcriptome raw data.The results of differential expression analysis indicated that there were 5881 differentially expressed genes in the mycelium of Changbaishan Polyporus umbellatus compared with the mycelium of Shaanxi Polyporus umbellatusof which there were 2616 up-regulated genes,and the down-regulated genes were 3265.The GO annotation classification of differentially expressed genes showed that the differentially expressed genes were mainly distributed in the membrane,membrane part,cell,cell part and organelle in the cell component;In the molecular function,the differentially expressed genes were mainly related to catalytic activity and binding;In the biological process,it is mainly related to metabolic process and cellular process.The KEGG classification of differentially expressed genes indicates that differentially expressed genes are mainly involved in carbohydrate metabolism,translation,amino acid metabolism,lipid metabolism,transport and catabolism,signal transduction,folding,sorting and degradation,etc.Pathway significant enrichment analysis results showed that nucleotide excision repair,base excision repair and plant-pathogen interaction metabolic pathways were all down-regulated in the Changbaishan Polyporus umbellatus mycelium;and Amino sugar and nucleotide sugar metabolism,Glycine,serine and threonine metabolism,Starch and sucrose metabolism,and Lysosome metabolic pathways were down-regulated in Changbaishan Polyporus umbellatus mycelium,indicating that the level of cell metabolism in Changbaishan Polyporus umbellatus mycelium was not as good as in Shaanxi Polyporus umbellatus mycelium is vigorous.By analyzing the metabolic pathways of Amino sugar and nucleotide sugar metabolism and Fructose and mannose,it was found that a total of 28 gene products are involved in the Amino sugar and nucleotide sugar metabolism pathways.There are 49 Unigenes related to them,of which 13 differential genes are involved in the metabolism of 8 gene products.Among them,2 up-regulated genes are involved in the synthesis of chitinase and phosphoglucose mutant enzyme;11 down-regulated genes encode chitinase,chitin synthase,hexosamine,and N-acetylglucosamine-6-Phosphodeacetylase,UTP-glucose-1-phosphate urate transferase,mannose phosphate mutant and GDP-L-fucose synthase.There are 28 Unigenes related to fructose and mannose metabolism,involving 20 gene products,of which 4 differential genes are involved in the metabolism of 8 gene products.Two up-regulated genes are involved in encoding aldehyde reductase and fructose diphosphate aldolase,class Ⅱ;two down-regulated genes encode mannose phosphate mutant and GDP-L-fucose synthase,respectively.
