Flower Color Genetics And QTL Mapping Of H.‘Datong Huanghua’× H.‘Lullaby’ F1 Population

Abstract/Summary:
Hemerocallis is one of the world’s most famous perennial flowers,with many varieties and colors.Genetic analysis of flower color and QTL mapping using Hemerocallis plants as research objects are of great significance for understanding the genetic law of flower color and molecular marker-assisted breeding of perennial plants.In this study,based on the research of the interspecific genetic map of Hemerocallis genus constructed earlier by the research group,using the flower color-related genes as a reference,the EST-SSR primers were developed and the map was encrypted.Eyes and petals were measured for color.The genetic effect of flower color in each part of the flower was analyzed,and the corresponding QTLs were detected.The main findings are as follows:(1)Screened 133 flower-related genes,and developed 112 pairs of EST-SSR primers,of which 15 pairs of primers are shown on the map.The encrypted map has a total of 121 sites,of which 4 new sites are added,and a total of 10 linkage groups.The total length is 1650.6c M,the average length between markers is 13.64 c M,the density of genetic map is between 7.8c M-19.6c M,the first linkage group has the most markers,with 24.Linkage groups 7 and 10 are the least marked,with 7 respectively.(2)The color difference meter was used to determine the L *,a *,b * values of different parts of the flowers,and their genetic analysis and correlation analysis were performed.The heritability is 33.4%~77.12%.The heritability of the L * value of the inner valve is the highest,the heritability is 71.22%,and the heritability of the outer valve eye is the lowest,and the heritability is 33.40%.Significant and extremely significant correlations existed between different parts of L *,different parts of a *,and different parts of b*.The outer valve a * has the best correlation with the eye a * of the outer valve,with a correlation coefficient of 0.988.(3)The QTL analysis of different index traits and the color of the inner and outer petals of the F1 population flowers showed that a total of 21 QTL loci were mapped,which could explain the phenotypic variation rate of 17.9%~27.7%.There are 10 QTLs with a phenotypic variation of more than 20%.The third linkage group has the most QTL loci,with 7 QTL loci and all of them located in the same interval.The interval size is 34.36 c M.Among them,the LOD value of 3.05 at qtl WBE3.1 is linked to the marker sau594,a difference of 2c M.The LOD value 3 of the qtl NHH8.1 locus of the 8th linkage group is linked to the marker sau752.Every 2nd and 6th linkage group points to 1 QTL locus.Three QTLs related to E * of the inner valve eye were located,which could explain that the site with the highest phenotypic variation rate was located in the first linkage group.To sum up,this paper carried on the genetic and QTL mapping analysis of flower color in interspecific hybrid populations of Hemerocallis plants,located the QTL loci of flower-color related traits,laid a theoretical foundation for the innovation of Hemerocallis germplasm resources,and also carried out Fine locating of different traits of Hemerocallis and molecular marker-assisted breeding provide theoretical references.
