Abstract/Summary:
Wool is one of the main products of the sheep industry and an important raw material for the wool textile industry.Wool is developed from the hair follicles,an accessory organ in the skin, and its bending is one of the important indicators of wool quality. In order to understand the relationship between the subunits of the NF-κB family(NFκB1,NFκB2,RELA,RELB,and REL)and the bending character of wool, and to select the subunits closely related to the bending character of wool.Merino semi-fine wool sheep and Local sheep were selected in the experiment. Two different breeds of sheep with large differences in wool bending were used as research objects. The relative expression of protein and m RNA of NF-κB subunits in the back skin of different breeds of sheep were detected by Western Blot and real-time quantitative PCR.By analyzing the correlation between the NF-κB subunits and the bending character of wool,we explored their effect on the formation of bending character of wool. The results showed that all subunits of the NF-κB family were expressed in the back skin of the Merino and Local sheep.Among them, except for RELA,the relative expression of protein and m RNA of other subunits in the back skin of the Merino were higher than Local sheep. In back skin samples of Merino, the relative expression of protein and m RNA of NFκB2 is the highest among each subunit of the NF-κB family, and it is significantly different from other subunits; In back skin samples of Local sheep, the relative expression of protein and m RNA of RELA is the highest among each subunit of the NF-κB family, and it is significantly different from other subunits. Conclusion: NFκB1,NFκB2,RELB,and REL are positively correlated with wool bending.Among them,NFκB2 was the most positively correlated, followed by NFκB1,RELB and REL are smaller;and RELA is significantly negatively correlated with wool bending. The results suggest that NFκB2 may play a major positive role in regulating the formation of wool bending character,NFκB1 may be an secondary positive regulation role;RELA may have a negative regulation effect on the formation of wool bending character; While RELB and REL may scarcely regulate the formation of wool bending character.
