In this test,the drought resistance of 20 common bean varieties was compared by morphological and physiological indicators.The physiological,biochemical and molecular mechanisms of the selected drought-resistant and sensitive varieties were determined.The main contents and results of this study are as follows:1、 Taking 20 common bean varieties in Shanxi Province as the object,the growth and physiological characteristics of common bean under drought stress were studied.The net photosynthetic rate,transpiration rate,stomatal conductance,chlorophyll content and relative leaf content were obtained by principal component analysis These indicators of water can be used as key indicators of drought resistance,and through these indicators,the drought-resistant variety ’ Hong zi ya ta jia ’ and the sensitive variety’Wang zhong wang jia beans ’ were selected.2、 10% PEG simulated drought stress treatment was carried out on the two varieties of ’ Hong zi ya ta jia ’ and’ Wang zhong wang jia beans ’,and the physiological and biochemical characteristics of the leaves and roots of the two were compared.The content of SOD,POD,CAT and other indicators of the frame was higher than that of the sensitive variety’ Wang zhong wang jia beans ’,and the change range was slower during the treatment.The soluble sugar content and soluble protein content of ’ Hong zi ya ta jia ’ are also higher than those of’ Wang zhong wang jia beans ’,In addition,the proline content of ’ Hong zi ya ta jia’ changed more slowly than that of’ Wang zhong wang jia beans ’.3、 The transcriptome data of two kidney bean varieties under 10% PEG simulated drought stress showed that there were 1361 differentially expressed genes in ’ Hong zi ya ta jia ’,and 1447 differentially expressed genes in’ Wang zhong wang jia beans ’Differential genes were analyzed through the KEGG pathway database and found that 10 significant metabolic pathways were involved in the ’ Hong zi ya ta jia’;9 significant metabolic pathways were involved in’ Wang zhong wang jia beans ’.Through the analysis of the GO function database,it was found that 111 significant GO items were involved in the ’ Hong zi ya ta jia ’,and 125 GO items were involved in the’ Wang zhong wang jia beans ’.Through analysis of transcription factors,a total of 487 differential genes were involved,of which b HLH transcription factors were the most,35,followed by ERF,MYB-related,B3 and other transcription factor families.Among the 9genes obtained through the above screening,the EIX1-2(XM_007143566.1)gene is the receptor protein;the CALM(XM_007144763.1)gene is calmodulin;the frm A,ADH5,adh C(XM_007141519.1)genes are S-Hydroxymethyl)glutathione dehydrogenase / alcohol dehydrogenase;CML(XM_007137421.1)calcium binding protein;SAUR(XM_007131669.1)and SAUR(XM_007131677.1)are SAUR family proteins;PXG(XM_007153204.1)It is a peroxidase;GH3(XM_007155808.1)gene belongs to the auxin-responsive GH3 gene family;XM_007143361.1 gene is not annotated in KEGG,annotated as oxalate-Co A ligase in the Uniprot database and belongs to a transcription factor G2-like family.Then these9 genes were verified by q RT-PCR,and the results were basically consistent with the transcriptome results.
