KAT6A,(Lysine Acetyltransferase 6A),a member of the MYST family of histone Acetyltransferases,affects many cellular processes through histone and nonhistone Acetylation,including gene transcription,cell Senescence,and Cardiac Septum Development Memory T cell diversity,maintenance of normal Hematopoietic stem cell,Kat6 a gene mutation led to a variety of congenital malformation,most individuals showed significant growth retardation,microcephaly and morphological abnormalities.These results suggest that KAT6 A gene plays an important role in growth and development.Previous laboratory studies have found three copy number variants in the sheep KAT6 A gene,and no studies of KAT6 A in sheep have been reported so far.Therefore,in this study,the type and distribution of copy number variation of KAT6 A gene in four sheep breeds(small tail Han sheep,Big Tail Han sheep,Hu sheep and Chaka sheep)were detected by q PCR,bioinformatics and the correlation analysis was made with the growth characters.At the same time,the structure and function of KAT6 A gene in sheep were analyzed by bioinformatics,which provided reference for further revealing the function of KAT6 A gene in sheep.The findings of this study are as follows:1.Reveals the type and distribution of KAT6 A gene copy number variation in four sheep breedsThe copy number types of the three copy number variant regions of the KAT6 A gene of 672 individuals of four sheep breeds were tested.The KAT6 A gene CNV was found to have three different copy number variants in the four sheep breeds: Loss,Normal and Gain.The Normal type of copy number variation in the Big Tail Han sheep and Chaka sheep was found.The main population accounted for 44.83% and 41.67% respectively;the copy number variation of CNV2 locus in small tail Han sheep,Big Tail Han sheep,Hu sheep and Chaka sheep was mainly Loss type,accounting for 37.37%,46.55%,38.97 % and37.15% respectively;CNV3 has the same ratio of Gain type and Loss type in small tail Han sheep,both of which are 38.42%.The copy number variation in big tail Han sheep is mainly Gain type,accounting for 34.48%,copying in Hu sheep and Chaka sheep the number of Loss types was dominant,accounting for 46.32% and 42.72% respectively.Combining the types of three CNV loci in each individual,25 CNV genotypes were found in the small-tail Han sheep,and 16 CNV genotypes were found in the big-tail Han sheep,which was found in the Hu sheep of the 23 CNV genotypes,27 CNV genotypes were found in Chaka sheep.Among the four sheep breed samples studied,only the CNV type of LGN combination was found in Chaka sheep.2.Found that the copy number of sheep KAT6 A gene was significantly associated with growth traits.In small-tail Han sheep(n=190),CNV1 copy number variation type of KAT6 A gene was significantly correlated with chest width(P<0.05),CNV2 copy number variation type was significantly correlated with chest width and chest depth(P<0.05),and CNV3 copy number variation type was significantly correlated with chest width and cannon bone circumference(P<0.05).In the big tail cold sheep(n=58),there were significant differences in body height,body slanting length,cross height and cannon bone circumference in the CNV3 CNV variant type of KAT6 A gene(P<0.05).In Hu sheep(n=136),CNV1 copy number variation type of KAT6 A gene was significantly correlated with body height and rump length(P<0.05),CNV2 copy number variation type was significantly correlated with chest girth(P<0.05),CNV3 copy number variation type was significantly correlated with body height,chest girth,cannon bone circumference and rump length(P<0.05).In Chaka sheep(n=288),CNV3 copy number variation type of KAT6 A gene was significantly correlated with body slanting length(P<0.05).The combination of three CNV loci with less than 5 individuals was removed,and the association analysis of three CNV locus combinations with sheep growth traits was performed.It was found that LLL and NLN were in chest width and tube traits in small-tail Han sheep.Significant effects(P<0.05);in Hu sheep,LLL,LNL,LNG,NLG,GLG,GNG,GGL and GGG types had significant effects on body height,body slanting length,chest circumference,cannon bone,rump length and body weight,respectively(P<0.05).In the Chaka sheep,although the differences between the individuals of different combinations are different,they are not significant.The above results indicate that the sheep KAT6 A gene copy number variation can be used as a molecular marker for sheep breeding.3.Bioinformatics analysis of KAT6 A gene in SheepBy using software to analyze the bioinformatics of sheep.The results showed that KAT6 A gene was a hydrophilic protein,GRAVY was-0.921,there were many phosphorylation sites and five transmembrane regions,which were mainly located in the nucleus The secondary structure of protein is composed of H15,PDH,RING,Pfam: Moz and Coiled Helix.These results laid the foundation for the functional study of the KAT6 A gene. |
