MADS-box Genes Differentially Expressed In Diverse Sex Types In Cucumber(Cucumis Sativus L.)

Abstract/Summary:
MADS-box gene family is a kind of transcription regulatory factors which affects plants’ growth and development,especially the development of flower organs.With the research of MADS-box genes in Arabidopsis and Antirrhinum,we have known how they work,and the ABCDE model of floral organ development is one of their functions.Cucumber is very important to people’s daily life,its production receives extensive attention all over the world.Cucumber’s production is decided by its sex-type,only female and hermaphrodite flowers can become fruits.MADS-box genes in cucumber could affect the ratio of male and female flowers,but the functions are not so clear.This research used six couples of NIL(near isogenic lines)materials for transcriptome analysis of different sex types of cucumber,including differential expression analysis,egg NOG analysis,GO enrichment analysis of differential expressed gene and KEGG enrichment analysis differential expressed gene.There are 9 MADS-box genes differentially expressing in floral buds and apical buds of different sex types: Cucsa.018420、Cucsa.113420、Cucsa.251170 and Cucsa.327970 expressed specifically in male flowers;Cucsa.139620、Cucsa.160640 and Cucsa.241990 differentially expressed in female flowers and perfect flowers;Cucsa.212720 and Cucsa.392160 expressed specifically in female flowers.In addition,in this research compares the differentially expressed MADS-box genes above were compared with other three plants’(Arabidopsis thaliana、Oryza sativa、Vitis vinifera L.)ABCDE classes genes.The results accorded with the transcriptome data.In the promoterregions of differential MADS-box genes,some transcription regulation elements were found to be related to sex determination of cucumber in promoter regions,such as G-box,ERE,these elements could affect sex-type of cucumber plants through plant hormone signals.Expression patterns of Cucsa.018420 and Cucsa.392160 were verified the same as what the transcriptome analysis showed.So this research discussed how MADS-box genes affected sex type of cucumber and it provides a reference to deeper study of MADS-box genes in cucumber.
