Experiment 1: Effect of Tremella fuciformis spore ferment polysaccharide on production performance and immune function of sow and offspring.The reproductive performance of sows is one of the most important parts in swine production,which is closely related to the economic benefit of Livestock farm.Polysaccharide has the effect of improving reproductive performance of sow and health of offspring,however,the study on the reproductive performance and offspring health of Tremella fuciformis spore ferment polysaccharide(TFP)has not been reported.A total of40 multiparous sows,with similar body weight(264±7.52 kg)and backfat thickness(15.71±0.94 mm),at parity(3~5)on 90 day of gestation were assigned to 2 dietary treatments randomly: control(CON),basal diet;and Tremella fuciformis spore polysaccharide(TFP),basal diet with 400 mg/kg TFP supplementation.20 replicates per treatment and 1 sow per replicate.The trial period was from d 90 of gestation to d 35 of piglets.On d 21 of age,11 sows were selected from CON and TFP,respectively.Piglets(two piglets per sow)were divided into two groups based on piglet diet supplemented with a TFP(0 or 400 mg/kg),resulting in four treatment groups:(1)CC(control sows–control piglets);(2)CT(control sows–treated piglets);(3)TC(treated sows–control piglets);(4)TT(treated sows–treated piglets).The colostrum,mature milk and weaning blood of sows,bloods of piglets in umbilical cord and at age of 21 d and 35 d,the tissue of duodenum,jejunum,ileum tissue were collected.The results showed that TFP addition in diet of sows was significantly increased the concentration of lactose(P<0.05)and ADG(P<0.01)of piglets during 15~21 d of lactation period.Sow dietary treatment tended to increase the levels of Ig M and Ig G(P< 0.1)in the plasma of piglets at 21 days old.Meanwhile,Piglets fed TFP diet had decreased faecal score(P<0.05)from d 1 to d 14 of trial.The m RNA abundance of TLR4(P<0.05),TLR9(P< 0.1),IL-1β(P< 0.1)and TNF-α(P<0.1)were increased compared with piglets fed CON diet.Meanwhile,The m RNA abundance of Claudin-1(P<0.05),ZO-1(P<0.05)and Occludin(P< 0.1)were increased compared with piglets fed CON diet.The results indicated that dietary supplement 400 mg/kg TFP improved ADG of suckling piglets and concentration of lactose of milk during gestation and lactation period.Meanwhile,dietary supplement 400 mg/kg TFP decreased diarrhea score and influence the gene expression of innate immunity and barrier function.Experiment 2: Effect of Tremella fuciformis spore polysaccharide on growth performance and immune function of weaning piglets.This study was used to determine the effects of Tremella fuciformis spore polysaccharide(TFP)on growth performance,biochemical and immunological parameters of weaned piglets under LPS challenge.A total of 72 male piglets(Duroc × Landrace ×Yorkshire,DLY),weaned at 21day(initial body weight 6.40±0.17 kg),were randomly allocated to 3 groups with 6 replicates and 4 piglets per-pen replicate,respectively.The piglets received control diet(CON)or control diet with 400 mg/kg TFP or 800 mg/kg TFP for a period of 28 days,respectively.On 29 d,two piglets were selected per-pen of CON and 400 mg/kg TFP groups to be injected with LPS(50 μg/kg of BW)or an equivalent amount of sterile saline,resulting in a 2 × 2 factorial arrangement of treatments.Blood samples were collected at 0 h and 3 h after LPS challenge.The results showed that the ADG(P<0.05)and ADFI(P<0.1)of piglets fed 400 mg/kg TFP diet was greater than piglets fed control and 800mg/kg TFP diets from d 1 to d 7.Piglets fed 400 mg/kg TFP increased ADG during d 8 to d 14 and ADFI during d 15 to d 21 compared with piglets fed control diet.Piglets fed 400 mg/kg TFP increased ADG compared with piglets fed control and 800 mg/kg TFP diets during the overall phase.Piglets fed 400 mg/kg TFP diet had decreased faecal score from d 1 to d 7 of trial(P<0.05)and during the overall phase(P<0.1)compared with piglets fed control diet.There was no significant effect on growth performance and faecal score of piglets fed 800 mg/kg TFP compared with piglets fed control diet.At 0 h after LPS challenge,piglets fed TFP diet had the higher plasma level of WBC(P<0.05)compared with piglets fed CON diet.At 3 h after LPS challenge,the level of WBC was significantly decreased,while the levels of RBC,HGB,HCT,TP,IL-1β and TNF-α(all P<0.05)were significantly increased of piglets challenged with LPS compared to piglets without LPS challenge.Meanwhile,the plasma concentrations of TP,IL-1β and TNF-α were decreased(all P<0.05)in piglets fed TFP compared with piglets fed CON diet.In conclusion,piglets fed 400 mg/kg TFP diet improved growth performance during first week after weaned,decreased diarrhea rate and improved the inflammatory response of piglets under acute immune challenge.However,there was no significant effect on the growth performance and immune function of piglets feed 800 mg/kg.
